Description

Method for simultaneously extracting eucommia male flower flavone and iridoid glycoside by using deep eutectic solvent and application thereof Technical Field The invention belongs to the technical field of substance extraction, and particularly relates to a method for simultaneously extracting eucommia male flower flavone and iridoid glycoside by using a deep eutectic solvent and application thereof. Background Eucommia ulmoides belongs to hermaphrodite tree species and is a unique precious medicinal plant in China. The flowers of the stamen plants are called eucommia stamen flowers (Eucommia ulmoides male flowers), and are approved as new food raw materials by the national health accouterment in 2014. In addition, various products such as tea, health care products, functional beverages and the like which take eucommia male flowers and extracts thereof as raw materials in the market are widely welcomed by consumers, and great attention is paid to the pharmaceutical and cosmetic industry. The eucommia male flowers not only have rich nutritive values, but also have remarkable health care effects of reducing blood pressure, blood fat, blood sugar and the like. In addition, it has anti-fatigue and anti-aging properties. Eucommia male flowers contain various bioactive components including iridoids (such as Geniposide (GP) and Aucubin (AU)), lignans (such as pinoresinol diglucoside), and flavonoids (such as isoquercitrin, rutin, and 3-arabinosyl quercetin). However, most extraction methods of eucommia male flowers still use conventional organic solvents, which constitute a certain risk factor for the processing of foods and cosmetics. Although water can replace the organic solvent, the extraction efficiency is too low. Therefore, an efficient and green extraction method should be developed to extract the target compound from the eucommia male flowers. Flavonoid and iridoid glycosides in eucommia male flowers have excellent biological activities such as oxidation resistance and skin photoaging resistance [ Liu Cong, guo Feifei, shongjunping, and the like, chemical components and pharmacological actions of different parts of eucommia are researched and developed [ J ]. Chinese journal of traditional Chinese medicine 2020,45 (03): 497-512 ], but the optimal condition for extracting flavonoid components in eucommia male flowers is ethanol concentration 60% [ Bai Xiting, zhu Wenxue, luo Lei, and the like, the extraction process research of flavonoid compounds in eucommia male flowers [ J ]. Food research and development, 2008, (11): 96-100 ], and the extraction condition of iridoid glycosides is 95% ethanol [ Ding Yanxia, wang Tengyu, zhang Yaowen, and the like, the triterpene chemical components in eucommia male flowers are researched [ J ]. Chinese journal of traditional Chinese medicine 2014,39 (21): 4225-4229 ], and the traditional organic solvent is difficult to extract simultaneously, and stable liquid dosage forms are obtained. Therefore, there is a need to develop a new green extraction solvent to achieve multi-active extraction and form a stable solvent. The invention aims to provide a liquid solvent which can simultaneously extract flavonoid and iridoid glycoside active ingredients and form stable antioxidant activity and anti-aging activity superior to those of organic solvent extracts. The research aims at exploring the possibility of extracting total flavonoids and iridoid glycosides from eucommia male flowers by using a deep eutectic solvent, and comparing with the extraction by using an organic solvent, whether the deep eutectic solvent can improve the concentration of the extracted flavonoids and iridoid glycosides, and has similar or stronger antioxidant and anti-aging activities. Disclosure of Invention Aiming at the defects of the prior art, the invention aims to provide a method for simultaneously extracting eucommia male flower flavone and iridoid glycoside by using a deep eutectic solvent and application thereof. In order to achieve the aim of the invention, the invention adopts the following technical scheme: In a first aspect, the present invention provides a method for simultaneously extracting eucommia male flower flavone and iridoid glycoside by using a deep eutectic solvent, the method comprising: (1) Pretreating eucommia male flowers, namely cleaning and deactivating enzyme of eucommia male flowers and drying; (2) Extracting eucommia male flower flavone and iridoid glycoside components by mixing pretreated eucommia male flower with solution containing deep eutectic solvent, heating to obtain crude extract, centrifuging the crude extract, and filtering.
