Abstract

The invention relates to the technical field of traditional Chinese medicine gel preparation, in particular to asiaticoside gel and a preparation method and application thereof. The asiaticoside gel is used in preparing wound healing preparation and consists of asiaticoside extract or liposome in the amount of 70-80wt% of the total asiaticoside as the active component 0.2-2 wt%, p-hydroxybenzoate compound 0.2-2 wt%, glycerin 2-10 wt% and carbomer 5-20 wt%. The preparation method comprises the steps of S1 swelling carbomer, adjusting pH to obtain carbomer gel, S2 dissolving asiaticoside extract or liposome and parahydroxybenzoate compound into glycerol to obtain glycerol solution, and S3 adding glycerol solution into carbomer gel, and stirring uniformly to obtain asiaticoside gel. The asiaticoside gel prepared by the invention has the advantages of no toxic or side effect, freshness, transparency, no greasiness and good wound healing effect in the wound healing process.

Description

Asiaticoside gel and its preparation method and application Technical Field The invention relates to the technical field of traditional Chinese medicine gel preparation, in particular to asiaticoside gel and a preparation method and application thereof. Background Centella asiatica is a plant of centella genus of Umbelliferae, widely distributed around the world, and mainly distributed around the south of Yangtze river basin in China. All herbs are used as medicines. The Chinese pharmacopoeia describes the functions and indications of the Chinese pharmacopoeia as clearing heat, promoting diuresis, removing toxin and relieving swelling. Can be used for treating jaundice due to damp-heat, heatstroke, diarrhea, stranguria with blood, carbuncle, sore, and traumatic injury. Centella mainly contains polyacetylene, triterpene and its glycosides, volatile oil, flavonoid, alkaloid and other compounds. Among them, asiaticoside, which is a triterpene, is considered as a main active ingredient of centella asiatica that exerts the skin-repairing effect. The asiaticoside compounds mainly comprise asiaticoside-B, madecassoside and asiaticoside. Asiaticoside can regulate inflammation, prevent excessive reaction, stimulate fibroblast proliferation and migration, and inhibit scar hyperplasia. The centella asiatica added into most products in the market at present has complex components, low asiaticoside content, undefined biotoxicity and fuzzy treatment effect. Disclosure of Invention In view of the technical problems identified in the background art, the object of the present invention is to prepare an asiaticoside gel and to carry out related cellular and animal tests on high asiaticoside contents to verify its toxicity and performance. The preparation method of the gel is simple to operate. The asiaticoside gel comprises carbomer, asiaticoside extract or liposome, glycerol, and parabens. The asiaticoside gel comprises, by weight, 0.2-2 parts of asiaticoside extract or liposome, 0.2-2 parts of parahydroxybenzoate compounds, 2-10 parts of glycerol, and 5-20 parts of carbomer. Wherein the centella asiatica total glycosides content in centella asiatica extract or liposome is 70-80wt%, and the parahydroxybenzoate compound is methyl benzoate, ethyl benzoate, butyl benzoate. The preparation method of the asiaticoside gel comprises the following steps: 1) Dispersing carbomer powder in water under stirring, standing overnight, fully swelling, adding triethanolamine or NaOH, and adjusting pH to 7-8 to obtain carbomer gel; 2) Adding the parahydroxybenzoate compound and asiaticoside extract or liposome with total asiaticoside content of 70-80wt% into glycerol to prepare glycerol solution; 3) Adding glycerol solution into carbomer gel, and stirring to obtain asiaticoside gel. The asiaticoside gel can be used for preparing gel for wound healing. The beneficial effects are that: the carbomer gel is used as a slow release agent, and the effective component asiaticoside is wrapped in the carbomer gel, if the asiaticoside liposome is adopted, the two are synergistic, so that the release speed can be more effectively and accurately controlled, the rapid loss of the effective component asiaticoside is avoided, and the asiaticoside is ensured to continuously act on the wound surface. After the asiaticoside liposome is smeared on skin, the phospholipid in the liposome and the lipid between cells of the horny layer are mutually fused, so that the lipid channel is enlarged. Gel and glycerol can help skin to be hydrated better, fully hydrated keratinocytes are swollen, cell gaps are increased, the three are combined, permeability is increased, asiaticoside can penetrate through the stratum corneum more easily, and transdermal effect is improved. The asiaticoside gel of the present invention has no toxic side effect, and is clear, transparent and oily, and has excellent wound healing effect.
