Abstract

The invention discloses an American ginseng fermentation broth with the function of resisting myocardial ischemia or dilating blood vessels and a preparation method and application thereof, wherein the preparation method comprises the following steps of 1) adding pure water into American ginseng, heating, refluxing and extracting, filtering, merging filtrate, and concentrating the filtrate to obtain concentrated solution; 2) mixing the concentrated solution with MRS culture medium containing lactobacillus plantarum, sealing and fermenting to obtain crude fermentation liquor, inactivating, centrifuging to obtain supernatant, and 3) concentrating the supernatant under reduced pressure at 50-60 ℃ to 1/3-2/3 of the volume of the supernatant to obtain American ginseng fermentation liquor. The American ginseng fermentation liquor PPD type and PPT type ginsenoside has more abundant types and stronger action foundation. Experiments prove that the American ginseng fermentation broth can improve the abnormal ejection fraction and ion exchange caused by myocardial ischemia, and has the effect of resisting myocardial ischemia. Experiments prove that the American ginseng fermentation broth can reduce blood pressure and has the function of dilating blood vessels.

  1. 1. A preparation method of American ginseng fermentation broth with the effect of resisting myocardial ischemia or dilating blood vessels is characterized by comprising the following steps: 1) Adding purified water into radix Panacis Quinquefolii, heating and reflux-extracting, filtering, mixing filtrates, and concentrating to obtain concentrated solution; 2) Mixing the concentrated solution with MRS culture medium containing lactobacillus plantarum with OD 600 value of 0.5-0.9 according to the volume ratio of 95-99:5-1, sealing and fermenting at 35-39 ℃ and 10-60 rpm for 24-48 hours to obtain crude fermentation broth, inactivating, centrifuging to obtain supernatant; the lactobacillus plantarum is Lactobacillus plantarum ATCC8014; 3) Concentrating the supernatant under reduced pressure at 50-60 ℃ to 1/3 to 2/3 of the volume of the supernatant to obtain the American ginseng fermentation broth.

The preparation method of the American ginseng is characterized by comprising the following steps of 1) adding purified water into American ginseng, heating and refluxing for extraction twice, wherein the mass of the added purified water is 8-12 times and 7-9 times of that of the American ginseng in sequence, extracting for 1.5-3 hours each time, filtering, combining the filtrates, and concentrating under reduced pressure at 50-60 ℃ to 4-6 times of that of the American ginseng to obtain concentrated solution.
  3. 3. The method according to claim 1, wherein the inactivating step comprises heating to 55-65deg.C for 25-35min, heating to 70-80deg.C, and holding for 3-10 min.

Thus, vasodilation and prevention of myocardial ischemia are key therapeutic approaches for cardiovascular disease. Research shows that American ginseng can obviously reduce the activities of myocardial phosphocreatine kinase, lactic dehydrogenase and aspartic acid amino-transferase of experimental myocardial infarction dogs, can obviously reduce the contents of myocardial lipid peroxide and free fatty acid, and can improve the activities of superoxide dismutase and glutathione peroxidase. However, due to weak potency of small molecular compounds such as American ginseng saponins, terpenes, hydrocarbons and the like, there are few related studies and patents on American ginseng to dilate blood vessels and resist myocardial ischemia.
