Description

Dandelion root extract, preparation method thereof and application thereof in promoting peripheral nerve regeneration Technical Field The invention relates to a Chinese herbal medicine extract, in particular to a dandelion root extract, a preparation method thereof and application thereof in promoting peripheral nerve regeneration. Background Herba Taraxaci (Dandelion, taraxacum) is a medicinal and edible Chinese herbal medicine, and has been widely grown around the world. The dandelion root is rich in various bioactive components such as polysaccharides, flavonoids, sterols, phenolic acids, choline, organic acids and the like, has bitter taste, sweet and cold nature, and is a traditional Chinese medicine for clearing heat, detoxicating, detumescence and resolving hard mass. At present, the research on dandelion and pharmacology thereof is focused on pharmacological effects of resisting viruses, resisting inflammation, resisting tumors, reducing blood sugar and blood fat and the like, and dandelion granules, capsules and the like are developed for treating clinical acute and chronic gastritis, sphagitis, sore and toxic swelling, canker sore and the like. Myelin is a unique anatomical structure of the peripheral nervous system, and neuronal electrical signals are conducted in jumps at the femtoknots of the myelinated nerve fibers Lang. Glial cells encapsulate the nerve axons to form myelin membranes, and myelin-associated glycoproteins (Myelin Associated Glycoprotein, MAG) synthesized and secreted by Schwann cells are located in the innermost myelin sheath, and are important transmembrane glycoproteins for myelin, in close proximity to axons emanating from neurons. Myelin basic protein (Myelin Basic Protein, MBP) is located on the myelin surface, 30% of the total myelin protein, a unique basic protein in the myelin sheath of the nervous system, and has a critical role in maintaining the stability of the nervous system. Myelin is a key component affecting the rate of nerve impulses transmitted to nerve fibers, and therefore, the restoration of myelin plays a decisive role in the effects of peripheral nerve regeneration. The effect of dandelion or its major components on the peripheral nervous system is not known. Disclosure of Invention The invention aims to provide dandelion root extract, a preparation method thereof and application thereof in promoting peripheral nerve regeneration. The preparation method of the dandelion root extract comprises the following steps of extracting acetone, triethanolamine and sodium borohydride from dandelion root. The acidic effective components in dandelion root include flavonoid, phenolic acid, organic acid and the like, the acetone has strong solubility to the flavonoid and the phenolic acid, and the triethanolamine alkaline auxiliary solvent can enable the flavonoid, the phenolic acid and part of alkaloid to be dissolved out more easily. Therefore, the combination of the acetone aqueous solution and the triethanolamine has the characteristics of good solubility and less impurities for the acidic active ingredients. In addition, sodium borohydride (NaBH 4) is used as a mild reducing agent, so that aldehyde and ketone impurities in the crude extract can be removed, and the easily-oxidized effective components in the crude extract are protected and stabilized. The volume ratio of the acetone to the triethanolamine is preferably 5% -95%, more preferably 55% -75%, 25% -45%, most preferably 65% -35%, and the concentration of sodium borohydride in the extracting agent is preferably 0.125-2.0 mol/L, more preferably 0.25-1.0 mol/L, most preferably 0.5 mol/L. The preparation method preferably comprises the following steps of (1) cleaning dandelion roots, baking and grinding into powder, (2) soaking the powder in an extractant, and (3) filtering, concentrating and freeze-drying the soaking solution to obtain dandelion root extract. In the step (1), the baking temperature is preferably 60-65 ℃. In step (2), ultrasonic-assisted extraction is preferably used. In the step (3), the concentration temperature is preferably 35-40 ℃, and most preferably 40 ℃. The invention also provides the dandelion root extract obtained by the preparation method. The dandelion root extract contains flavone and sterols. The invention also provides application of the dandelion root extract in promoting peripheral nerve regeneration. The use is preferably for the preparation of a medicament for peripheral nervous system disorders. Compared with the prior art, the invention has the following remarkable advantages: 1. The dandelion root extract provided by the invention is rich in flavone and sterol substances, can promote the growth of nerve cells and neurites, promote the increase of mRNA expression of myelin-associated factors, promote the morphological structure regeneration and functional recovery of myelin sheath layers of damaged nerves, and can be used for repairing peripheral nerve injuries and promoti
