Abstract

The invention discloses a compound traditional Chinese medicine aqueous extract for preventing and treating infectious bronchitis of chickens, a preparation method and application thereof, and belongs to the field of veterinary traditional Chinese medicine preparations. The water extract is prepared from six medicinal materials of rhizoma pinellinae praeparata, fritillaria thunbergii, dried orange peel, liquorice, platycodon grandiflorum and bitter apricot seed according to a specific proportion, and is prepared by water extraction and concentration. Aiming at the defects of the existing control scheme in recovering the growth performance and immune function of infected chickens, the invention creatively adopts a therapeutic formula combining 'regulating qi, strengthening spleen, drying dampness, resolving phlegm' and 'dispersing lung qi'. Animal experiments show that the compound can not only effectively inhibit the replication of infectious bronchitis viruses of chickens, but also remarkably improve thymus indexes of infected chickens, and enable average daily gain of the chickens to be recovered to a level which is not remarkably different from that of a healthy group, and the synergistic effect is obviously higher than that of a ribavirin positive control group. The invention has simple components, stable process and good safety, and provides a new product for the green and efficient prevention and treatment of infectious bronchitis.

Description

Traditional Chinese medicine compound water extract for preventing and treating infectious bronchitis of chickens, and preparation method and application thereof Technical Field The invention belongs to the technical field of veterinary traditional Chinese medicine preparations, and in particular relates to a traditional Chinese medicine compound aqueous extract for preventing and treating Infectious Bronchitis (IB), a preparation method thereof and application thereof in preparing veterinary medicines. More specifically, the invention relates to a six-ingredient traditional Chinese medicine compound with the synergistic effect of antivirus-immunoregulation-growth recovery, which can effectively inhibit replication of Infectious Bronchitis Virus (IBV) of chickens and remarkably improve development and production performance of immune organs of infected chickens. Background IB is an acute, highly contagious respiratory infectious disease caused by IBV of the coronaviridae family. The disease spreads rapidly, the serotypes are complex and easily changed, the vaccine immune protection is incomplete, and the continuous economic loss is caused for the global poultry industry. In clinical treatment, chemical synthesis of antiviral drugs such as amantadine, ribavirin and the like has been definitely prohibited from being used in edible animals due to strict requirements of food safety and public health, so that the breeding industry faces serious challenges of limited therapeutic drug selection。 Under the background, the traditional Chinese medicine compound with the advantages of multiple components, multiple targets and low residue becomes an important research direction for preventing and treating the virus diseases of livestock and poultry. At present, the traditional Chinese medicine compound for preventing and treating chicken IB is disclosed and reported and clinically used, and the formula treatment is mainly focused on dispersing lung and clearing heat or relieving cough and asthma, and the main aim is to directly relieve clinical symptoms of respiratory tract. The classical prescription of Maxingshigan decoction is widely applied in modern veterinary clinic and research, and is used as a common positive control for evaluating related drug effects. The proposal has certain effect on improving exterior symptoms such as cough, asthma and the like. However, the applicant has found through long-term clinical observations and experimental studies that the pathological processes of IBV infection are far beyond the pulmonary system. After invasion of the virus, the lung qi is easy to be blocked, and the disorder of the dispersing and descending is easy to be caused, so that the primary and secondary diseases seriously affect the transportation and transformation functions of the spleen and stomach. Spleen and stomach are the acquired sources of qi and bloodThe direct consequence of dysfunction is that first, water-dampness stops internally, gathers to generate phlegm, and forms a secondary pathological product of 'phlegm-dampness', which further hinders qi movementThe second, qi and blood biochemical source of weakness leads to deficiency of healthy qi, which is manifested by significant inhibition of development of central immune organsThirdly, the organism nutrition supply and utilization disorder causes serious and continuous growth arrest of infected birds. This constitutes a deep pathological chain of "lung failing to disperse and descend → spleen failing to transport → phlegm-dampness internal resistance → immunosuppression & growth arrest". The existing control strategy mainly used for clearing and dispersing lung heat has the action center of lung, and has limited effects on the systemic problem of immune suppression and growth recovery which is derived from spleen and affects the whole body. The method directly leads to a common phenomenon in clinic that after respiratory symptoms of infected chickens are relieved by the prior proposal, indexes of immune organs such as thymus and the like are slowly recovered, cellular immunity level is low, average daily gain cannot be recovered to a healthy level for a long time, growth recovery period is prolonged, and finally, the economic benefit of cultivation is severely restricted. Therefore, the industrial practice is urgent to develop an innovative traditional Chinese medicine compound capable of directly striking the heart disease machine of spleen dysfunction transportation and internal stagnation of phlegm-dampness on the basis of inheriting the traditional antiviral advantages of traditional Chinese medicines and breaking through the conventional therapeutic principle of mainly aiming at lung and heat, thereby systematically solving the difficult problem of cooperative recovery of immunosuppression and growth stagnation. In the prior art, although a plurality of traditional Chinese medicine formulas for treating respiratory diseases exist, the traditional
