Abstract
The invention particularly discloses application of a starch-astragalus polyphenol compound in preparation of a regulating and controlling preparation for improving cognitive function decline, wherein the starch-astragalus polyphenol compound is a compound with a V-shaped crystal structure formed by carrying out alcohol-water system damp-heat treatment on starch and astragalus polyphenol or derivatives thereof. The invention discloses that the compound can be used for regulating and controlling the intestinal short-chain fatty acid generation and neurotransmitter metabolic balance through double-target-point synergy for the first time, thereby improving the cognitive function reduction induced by factors such as high-fat diet and the like. Compared with free stilbene polyphenol or physical mixture, the invention has strong targeting and good and stable cognitive function intervention effect.

- 1. The application of the starch-astragalus polyphenol compound in preparing a regulating preparation for improving cognitive function decline is characterized in that the starch-astragalus polyphenol compound is a compound with a V-shaped crystal structure formed by carrying out alcohol-water system damp-heat treatment on starch and astragalus polyphenol or derivatives thereof.

The use according to claim 1, characterized in that the preparation method of the starch-stilbene polyphenol complex comprises the following steps: (1) Regulating the starch raw material to 5-35 wt% of moisture content, and standing to achieve moisture absorption balance; (2) Adding stilbene polyphenol or a derivative thereof into the starch, wherein the addition amount of the stilbene polyphenol or the derivative is 1-10 wt% of the dry mass of the starch; (3) Adding ethanol into the solid phase mixed system in the step (2), wherein the addition amount of the ethanol is 5-30 wt% of the mass of the dry starch, uniformly mixing, and reacting 1-5 h in a closed container at 70-150 ℃ to enable starch chains to be partially rearranged and embedded into stilbene polyphenol or derivatives thereof to form a V-shaped crystal structure; (4) Washing after the reaction is finished, drying, crushing and sieving to obtain the starch-astragalus polyphenol compound.
- 3. The use according to claim 2, wherein the moisture content in the step (1) is controlled within the range of 20+/-5 wt%, and the ethanol addition in the step (2) is 20+/-5 wt% of the dry mass of the starch.
- 4. The use according to claim 2, wherein the reaction temperature in step (3) is 90-130 ℃ and the reaction time is 1-3 h.
- 5. The method according to claim 2, wherein the standing temperature in the step (1) is 25.+ -. 10 ℃ and the time is 24.+ -. 12 h, the solvent used for the washing in the step (4) is 20-80% (v/v) ethanol-water solution, the drying temperature is 30-60 ℃ and the drying time is 8-24 h.

The use according to any one of claims 1 to 5, wherein the starch-stilbene polyphenol complex is selected from resveratrol, resveratrol glycoside, oxidized resveratrol, resveratrol triacetate, piceid, pterostilbene, polygonin or a mixture thereof, and the derivative of stilbene polyphenol comprises an esterified substance, an etherified substance or a glycosidated substance thereof.
- 7. The use according to any one of claims 1 to 5, wherein the starch is selected from one or more of corn starch, rice starch, wheat starch, tapioca starch and potato starch.
- 8. The use according to any one of claims 1 to 5, wherein the amount of stilbene polyphenol or derivatives thereof added to the starch-stilbene polyphenol compound is 3-9 wt% of the dry mass of the starch.

The use according to any one of claims 1 to 5, wherein the starch-stilbene polyphenol complex acts by dual-target modulation of intestinal brain axis chemical signals, the modulation formulation comprising a drug for improving cognitive dysfunction and a functional food or nutritional supplement for promoting cognitive functional health.
- 10. The use according to claim 9, wherein the starch-stilbene polyphenol complex regulates intestinal short-chain fatty acid production and neurotransmitter metabolic balance by means of dual-target synergy, improving or promoting animal performance in learning, memory or cognitive behaviours.

Application of starch-astragalus polyphenol compound Technical Field The invention belongs to the field of food science and nutrition intervention, and particularly relates to application of a starch-astragalus polyphenol compound in a regulating preparation for regulating intestinal and brain axis chemical signals to improve cognitive function decline. Background Cognitive dysfunction is manifested by a decline in the multidimensional abilities of learning, memory, attention and executive function, which occurs in close association with high-fat diet, metabolic disorders, chronic low-grade inflammation and intestinal microecological imbalance. In recent years, it has been found that the gut brain axis (gut-brain axis) plays a key role in the maintenance of cognitive function homeostasis as an important bi-directional regulatory pathway connecting gut microorganisms to the central nervous system. The substances have oxidation resistance, inflammation resistance and neuroprotection potential, but have poor water solubility, low stability and limited bioavailability, so that the application effect of the substances in the field of dietary intervention is limited. Starch, as a safe, edible natural high molecular carbohydrate, can form stable complex structures with hydrophobic small molecules under specific conditions, thereby imparting superior gastrointestinal stability and colonic delivery characteristics to the active. In the prior art, there have been studies on the formation of complexes of starch with polyphenols (including part of stilbene polyphenols) to improve their solubility, stability or antioxidant activity in vitro. However, these studies have focused mainly on structural characterization of the complex, in vitro digestion properties, or general enhancement of antioxidant properties, and have not revealed in depth its specific targeting mechanism under complex physiological environments. In particular, aiming at how the starch-astragalus polyphenol compound passes through a specific channel of the intestinal brain axis, the double signal channels of 'short chain fatty acid generation' and 'tryptophan metabolism' are synchronously regulated and controlled, so that the cognitive function is synergistically improved, and no clear purpose design and systematic efficacy verification are available at present. Disclosure of Invention The invention aims to provide an application of a starch-astragalus polyphenol compound, and discloses that the compound can be used for regulating and controlling intestinal short-chain fatty acid generation and neurotransmitter metabolic balance through double-target-point synergy for the first time, so that cognitive function reduction induced by factors such as high-fat diet and the like is improved.
