Abstract

The invention discloses a preparation method and application of a chlorogenic acid-zein nanoparticle-loaded chitosan film, and belongs to the technical field of food packaging. The method comprises the steps of firstly preparing chlorogenic acid-zein nano particles by taking zein as a wall material through an anti-solvent precipitation method, then blending the chlorogenic acid-zein nano particles with chitosan film-forming solution, and performing tape casting film formation to prepare a composite film, wherein the technical problems of easiness in oxidative degradation of chlorogenic acid, poor water solubility, insufficient mechanical property and single function of a pure chitosan film are solved through regulating and controlling the nano particle preparation process and film composite parameters. The prepared composite film has excellent barrier property, mechanical property, thermal property and oxidation resistance, and can effectively delay the oxidative deterioration of millet in the storage period.

The preparation method of the chlorogenic acid-zein nanoparticle-loaded chitosan film is characterized by comprising the following steps of: (1) The preparation of chlorogenic acid-zein nano-particles comprises the steps of dissolving zein and chlorogenic acid in ethanol water solution for 30min to prepare zein and chlorogenic acid solution, mixing the prepared zein and chlorogenic acid solution, stirring and dissolving for 1h in a dark place, adding a certain volume of deionized water into the mixed solution, continuously stirring for 1h, and removing ethanol by rotary evaporation to prepare the chlorogenic acid-loaded zein nano-particle suspension. (2) The preparation of the chitosan film forming liquid comprises the steps of dissolving chitosan in acetic acid solution, adding glycerol, heating and stirring for 30min until the chitosan film forming liquid is completely dissolved, and cooling to room temperature to obtain the chitosan film forming liquid. Background With the increasing awareness of consumers on food safety quality and environmental protection, the development of novel food packaging materials which are biodegradable and have the function of delaying food deterioration has become a research hotspot in the fields of food science and material engineering. Therefore, the packaging film with the functions of biodegradability and food deterioration delay is an ideal solution for replacing the traditional plastic and improving the shelf life of the food. As natural aminopolysaccharide, chitosan (CS) has good film forming property, biocompatibility, degradability and certain antibacterial property, is an ideal base material for food packaging, but the pure chitosan film has the defects of insufficient mechanical property, limited barrier property and single function, and limits the application of the pure chitosan film. In order to impart antioxidant and antibacterial activities to packaging films, natural polyphenol compounds such as chlorogenic acid (CGA) are often added to the film matrix. Chlorogenic acid is a natural polyphenol active ingredient and has excellent antioxidant and antibacterial activities, but has extremely unstable chemical properties, is easily subjected to oxidative degradation under the influence of light and heat, has poor water solubility and low bioavailability, and is difficult to realize long-acting and stable activity exertion when being directly added into a film. Zein (Zein) is a natural plant protein, has good biocompatibility, degradability and hydrophobicity, is an ideal wall material for encapsulating hydrophobic/amphiphilic active ingredients, can be prepared into nano particles by an anti-solvent precipitation method to physically protect the active ingredients and improve the stability of the active ingredients, but no report on the preparation of functional films by compounding chlorogenic acid-Zein nano particles with chitosan exists at present. Therefore, it is necessary to develop a nanoparticle with natural zein as a wall material for encapsulating chlorogenic acid, and to prepare a functional film with high stability, high activity and excellent mechanical properties by compounding with chitosan, thereby helping to delay oxidative deterioration of millet in the storage period.
