Description
Bacillus desert B-3 with broad-spectrum antibacterial activity and application thereof Technical Field The invention belongs to the field of biological agriculture, and relates to bacillus desert B-3 with broad-spectrum antibacterial activity and application thereof. Background Soil-borne plant diseases form a continuous threat to agricultural production, wherein clubroot caused by brassica radiata is particularly prominent, and the management of the clubroot is called as a worldwide difficult problem. The dormant spores of the pathogenic bacteria can survive for more than ten years in the soil, and the transmission paths are various, so that once the field is polluted, the dormant spores are extremely difficult to eradicate. Currently, chemical pesticides have little effect on dormant spores of this bacterium and are difficult to act on pathogens that have invaded plant roots. Although the soil fumigation and other powerful chemical methods have certain effects, the cost is high, the operation is dangerous, the soil micro-ecology is seriously destroyed, and the method runs counter to the sustainable development of agriculture. Faced with this serious challenge, microbial antagonists are considered a promising green control approach. However, the biocontrol strains of the prior art often show significant limitations when dealing with plasmodiophora. Many strains have a narrow bacteriostasis spectrum and cannot simultaneously cope with secondary diseases caused by clubroot. More importantly, the acid or barren soil environment in which clubroot often occurs can severely restrict the survival, colonization and activity of most biocontrol bacteria, so that the field control effect is unstable, and the laboratory effect is difficult to reproduce. Thus, there is a great need in the art for a biocontrol microbial resource that is specifically adapted to and effective against the unique and persistent pathogen, plasmodium rhizomatous. The ideal new strain should be capable of effectively colonising in the severe soil environment where the clubroot bacteria live, and have broad-spectrum antagonistic activity to cope with compound infection, thereby providing a brand new solution for overcoming the chronic clubroot. The desert bacillus is an important member of bacillus, and is gradually deepened in research and application in the field of plant protection due to the characteristics of strong stress resistance, rich metabolites and the like. The disclosed research shows that the strain has proved to have inhibition effect on a plurality of plant pathogenic bacteria such as fusarium pseudocladi, fusarium graminearum, cotton fusarium wilt and the like, can realize disease prevention and control through mechanisms such as synthesis of antibacterial compounds, competition of ecological niches, induction of plant system resistance and the like, and has the multifunctional characteristics of promoting plant growth, degrading agricultural wastes and the like. The culture conditions are standardized step by step, the detection technology is optimized continuously, a foundation is laid for large-scale application, but up to the present, no public report has been made to confirm that the bacillus desert has the function of preventing and treating clubroot, and targeted strain screening and application research is lacking. Disclosure of Invention The invention aims at providing the bacillus desert B-3 with broad-spectrum antibacterial activity and the application thereof, aiming at the defects of the prior art. The invention also aims to provide a microbial inoculum, supernatant fluid and application thereof prepared by the bacillus desert B-3. It is still another object of the present invention to provide a method for controlling clubroot of cruciferous crops. The aim of the invention can be achieved by the following technical scheme: The bacillus desert (Bacillus inaquosorum) B-3 is preserved in China general microbiological culture Collection center (China center for type culture collection) with a preservation date of 2025, 8 and 28 days and a preservation number of CGMCC No.35756. The desert bacillus (Bacillus inaquosorum) B-3 is separated from eggplant stalks, is an opaque yellowish white circular colony on an LB plate, can grow on LB, PDA and YPDA culture mediums, has an optimal growth temperature of 28 ℃, and can produce protease and cellulase.
