Abstract

The invention discloses a common wild rice OrLRR gene and application thereof in improving rice blast resistance of rice, belonging to the technical fields of genetic engineering and plant pathology. The nucleotide sequence of the common wild rice OrLRR gene is shown as SEQ ID NO. 1. The OrLRR gene has important function in resisting rice blast. The OrLRR gene deletion reduces the resistance of rice to Pyricularia oryzae, while over-expression of OrLRR gene can improve the resistance of rice to Pyricularia oryzae. The invention provides an application of OrLRR gene or its coding protein in improving rice blast resistance and a method for cultivating rice blast resistant rice by regulating OrLRR1 expression. The invention provides new gene resources and theoretical basis for cultivating new rice varieties with high rice blast resistance, and has important application value.

Description

Common wild rice OrLRR gene and application thereof in improving rice blast resistance of rice Technical Field The invention belongs to the technical field of genetic engineering and plant pathology, and particularly relates to a common wild rice OrLRR gene and application thereof in improving rice blast resistance of rice. Background A fungal disease, which is caused by rice blast bacteria (Magnaporthe oryzae) and damages the growth of rice for a long time, occurs in the north-south rice areas of China every year. The rice blast is used as a main disease in rice production, has the characteristics of strong popularity, large occurrence area, serious hazard and the like, and can lead to 10% -35% yield reduction of the rice in the whole growth period of the rice throughout the year. Compared with chemical prevention and control modes and the like, the identification and utilization of the rice blast resistance gene from rice germplasm resources is the most economical and environment-friendly strategy for preventing and controlling rice blast. The rice blast resistance R genes or loci identified from cultivated rice are numerous but rarely have durable, broad-spectrum resistance. The vertical resistance of a single R gene is difficult to cope with physiological race diversity, more than 60% of existing main cultivated varieties only contain 1-2 resistance sites, and the genetic linkage of disease resistance and agronomic characters leads to low breeding selection efficiency. Although resistance breakthrough can be delayed by gene polymerization techniques, the growth inhibition effect induced by the superposition of multiple genes limits practical applications. Common wild rice (Oryza rufipogon griff.) is an ancestral species of rice, harboring a large number of excellent genes lost during domestication of rice, particularly at gene sites related to biotic and abiotic stress resistance. The novel disease-resistant QTL or gene is developed and utilized from abundant wild rice germplasm resources, the gene resources for disease-resistant breeding of cultivated rice are expanded, and the efficient broad-spectrum environment-friendly disease-resistant variety is cultivated, so that the method is an important way for breaking the bottleneck of disease-resistant breeding of rice. Disclosure of Invention The invention aims to provide a novel application of a common wild rice OrLRR gene in regulating and controlling rice blast resistance, and provides theoretical basis and technical support for cultivating a novel rice variety with high rice blast resistance. The research shows that the common wild rice OrLRR gene plays an important role in the rice blast resistance process of the rice. The OrLRR gene deletion reduces the basic defense capability of rice against Pyricularia oryzae, and over-expression of OrLRR gene can improve the disease resistance of rice against Pyricularia oryzae. In order to achieve the aim of the invention, the invention is realized by the following technical scheme: The invention provides application of a common wild rice OrLRR gene in improving rice blast resistance of rice, wherein the nucleotide sequence of the OrLRR gene is shown as SEQ ID NO. 1. The invention also provides a breeding method for improving the rice blast resistance of the rice, which comprises the step of improving the expression quantity of OrLRR genes in the rice. Further, the improvement of the expression level of OrLRR gene in rice is realized by introducing recombinant vector into rice. Further, the recombinant vector is obtained by inserting a nucleotide sequence shown as SEQ ID NO. 1 between restriction enzyme KpnI recognition sites of a WMV023 vector. Further, the rice is a rice variety of Nippon or Zhonghua 11. Advantageous effects The invention discovers that the common wild rice OrLRR gene plays a key positive regulation role in the rice blast defense, and the knockout of OrLRR gene leads to the remarkable reduction of the resistance of rice to rice blast. The invention obviously improves the resistance of the rice to rice blast by over-expressing OrLRR genes in the rice, provides a new gene resource for cultivating a new rice variety resistant to the rice blast, has simple and convenient operation, can be directly applied to rice breeding, and has important application value.
