CN-121975977-A - Nucleic acid molecule combination of pathogen standard and application thereof
Abstract
The invention belongs to the technical field of sequencing, and particularly relates to a nucleic acid molecule combination of pathogen standard substances and application thereof. The nucleic acid molecule combination of the pathogen standard can monitor multiple pathogens simultaneously, and can avoid sample pollution to generate false positive results by means of letter rejection, so that the problems that the existing pathogen standard can not completely monitor the detection level of a product to a target pathogen by only using one or more known pathogens are solved, but the type of PTC pathogen is easily polluted when the type of the PTC pathogen is increased.

Description
Nucleic acid molecule combination of pathogen standard and application thereof Technical Field The invention belongs to the technical field of sequencing, and particularly relates to a nucleic acid molecule combination of pathogen standard substances and application thereof. Background Recently, metagenomic second-generation high-throughput sequencing technology (mNGS) and pathogen microorganism-targeted sequencing (tNGS) have begun to be widely used in many research and clinical fields, which utilize sequencing and alignment of nucleic acids to identify the vast majority of microorganism species in a sample. During the application, each of mNGS and tNGS is tested using a negative control (NTC) and a positive control (PTC) to monitor the presence of anomalies during the test to ensure the reliability and stability of the test results. Typically, NTCs will use a pathogen-free nucleic acid of human genome, the test results of which can be used to monitor the reagent background and environmental background during the course of this run of experiments, to distinguish responsible from background pathogens in the sample batch. PTC typically incorporates a known pathogen or pathogen nucleic acid into a human genomic nucleic acid that is free of the pathogen nucleic acid, and the test results can be used to monitor whether abnormalities occur during this batch of experiments, resulting in undetectable positive pathogens. The pathogen in the PTC may be in the form of inactivated bacteria to mimic the actual infection of a bacterial fluid, or fragmented genomic nucleic acid to mimic the status of pathogen free nucleic acid (e.g., cfDNA) in a sterile fluid. Currently, commonly used PTC generally employ one or more known clinically pathogenic pathogens, such as acinetobacter baumannii, staphylococcus aureus. Part of the positive standard substance also has probiotics as PTC, so that pathogens in the positive standard substance are prevented from polluting the sample to be tested, and the interpretation is prevented from being correctly reported. Therefore, the number of pathogens in the PTC used at present cannot be too large, one is to avoid cross contamination between samples in the same batch, and the other is to avoid laboratory environmental pollution caused by long-term use of PTC in the laboratory, which results in sample contamination between laboratory batches. However, the performance of the mNGS and tNGS products of different companies is currently uneven, and the detection level of the target pathogen of the products cannot be completely monitored by only using one or more known clinical pathogenic pathogens, especially for monitoring the detection actual conditions in the pathogen spectrum of tNGS products. But can easily cause contamination problems for increasing the variety of PTC pathogens. In addition, for a complete PTC, its function requires monitoring the entire process of mNGS and tNGS, including extraction, pooling, hybridization and sequencing. The extraction and monitoring of the bacteria liquid is also a difficult problem, because different pathogens are cultured in different modes, the preparation of different pathogens and the simultaneous incorporation of PTC (Positive temperature coefficient) preparation are time-consuming and labor-consuming, and some pathogens are often aggregated together or have volume differences, so that the quantity of the incorporated pathogens cannot be controlled accurately. Therefore, PTC of a variety of pathogens can be prepared without contamination problems, easy to operate and low cost, and the difficulty of accurately controlling the number of copies of pathogen standard is great.
