Abstract

The invention discloses application of bile acid in preparing a medicine for treating tumor cachexia. The invention provides and verifies that the bile acid, especially deoxycholic acid in secondary bile acid, can effectively restore the body weight of a tumor cachexia model mouse, increases the fat droplet size of fat cells and the weight and cross section area of fat tissues by inhibiting the level of protein related to fat degradation, and provides a therapeutic drug with great application potential for the symptoms of body weight loss and fat tissue exhaustion caused by tumor cachexia.

  1. 1. An application of bile acid in preparing medicine for treating tumor cachexia is provided.
  2. 2. The use of claim 1, wherein the bile acid is a secondary bile acid.
  3. 3. The use according to claim 2, wherein the secondary bile acid is deoxycholic acid.
  4. 4. The use according to claim 1, for the preparation of a medicament for improving the body weight of a patient suffering from tumour cachexia.
  5. 5. The use according to claim 1, characterized in that it is the use in the preparation of a medicament for the treatment of the symptoms of cancer cachexia with respect to the exhaustion of fat.
  6. 6. The use according to claim 5, wherein the symptom of fat depletion comprises fat breakdown, white adipose tissue browning.
  7. 7. The use according to claim 1, wherein the medicament contains bile acid or a pharmaceutically acceptable salt, solvate, hydrate, metabolite thereof as active ingredient.
  8. 8. The use according to claim 7, wherein the medicament further comprises pharmaceutically acceptable excipients.
  9. 9. The use according to claim 8, wherein the pharmaceutically acceptable excipients comprise any one or more of excipients, diluents, lubricants, glidants, wetting agents, emulsifiers, pH buffering substances, solubilizers, co-solvents or solvents.

The use according to claim 1, wherein the pharmaceutical dosage form comprises a tablet, capsule, granule, powder, chewable tablet, effervescent tablet, sustained release tablet, pellet, injection, infusion solution, suspension, patch, suppository, transdermal patch, microemulsion liposome, nanoparticle.

Description

Application of bile acid in preparation of medicine for treating tumor cachexia Technical Field The invention relates to tumour cachexia, in particular to application of bile acid in preparing a medicament for treating tumour cachexia. Background Tumour cachexia is a multifactorial metabolic disorder syndrome caused by tumours and is mainly manifested by progressive, involuntary weight loss, especially by sustained consumption of skeletal muscle and adipose tissue, often accompanied by anorexia, fatigue and systemic inflammatory responses. Among these, fat breakdown is one of the core features of cachexia, and rapid systemic fat loss occurs in patients even if they ingest sufficient calories, as opposed to the fat consumption pattern in normal starvation conditions. Fat degradation not only leads to the exhaustion of energy reserves, but also releases a large amount of free fatty acids, further exacerbating insulin resistance, liver fat infiltration and systemic low-grade chronic inflammation, forming a vicious circle of mutually exacerbated metabolic imbalance and tissue consumption. In addition, adipose tissue is not only an energy storage organ, but also has endocrine functions, and abnormal decomposition of adipose tissue can influence secretion of adiponectin, leptin and other fat factors, thereby indirectly participating in regulation of appetite and energy metabolism. At present, the treatment means for tumor cachexia are limited and have different effects. The main strategies include nutritional support, drug therapy and anti-tumor therapy. The progesterone derivative can be used for stimulating appetite, but can bring side effects such as thrombus, and the glucocorticoid can improve appetite and physical strength in a short term and accelerate muscle consumption in a long term. In general, existing therapies focus on relief of symptoms and fail to effectively reverse fat consumption, the root cause of which is that cachexia involves multi-pathway interactions, the mechanisms of which have not yet been fully elucidated, and in particular specific signal-regulating networks for fat degradation have yet to be explored further, limiting the development of targeted therapeutic strategies. Disclosure of Invention The invention aims to provide an application of bile acid in preparing a medicine for treating tumor cachexia, in particular to a medicine for treating the symptoms of weight loss and adipose tissue exhaustion of a patient suffering from tumor cachexia. The technical scheme is that the bile acid is applied to the preparation of the medicine for treating tumor cachexia. Preferably, the bile acid is a secondary bile acid, and more preferably, the secondary bile acid is deoxycholic acid. Preferably, the use is in the manufacture of a medicament for improving the body weight of a patient suffering from tumour cachexia. Preferably, the application is the application in preparing a medicine for treating the symptom of the exhaustion of the adipose tissue of the tumour cachexia, and further preferably, the symptom of the exhaustion of the adipose tissue comprises lipolysis and brown stain of the adipose tissue. Preferably, the medicament contains bile acid or a pharmaceutically acceptable salt, solvate, hydrate, metabolite thereof as an active ingredient. Compared with the prior art, the invention has the advantages that bile acid, especially deoxycholic acid in secondary bile acid, can effectively restore the body weight of a tumor cachexia model mouse, and can improve the weight and cross-sectional area of adipose tissue and increase the size of fat cell lipid drops by inhibiting the level of protein related to fat degradation, thereby providing a therapeutic drug with great application potential for the symptoms of weight loss and adipose tissue exhaustion caused by tumor cachexia.
