Abstract

The invention discloses application of Osmunda japonica in preventing and treating ichthyophthiriasis of aquaculture animals, and experiments on toxicity of Osmunda japonica to grass carp kidney Cells (CIK) show that the maximum non-toxic concentration of Osmunda japonica aqueous extract to CIK cells is 1200 mug/mL, the inhibition of Osmunda japonica aqueous extract to the survival rate of ichthyophthiriasis is dose-dependent, and when the drug concentration reaches 100 mg/L (mug/mL), the death rate of the grazing body reaches 100%, and in addition, the half-killing concentration (LD 50) of the Osmunda japonica aqueous extract to the grazing body of ichthyophthiriasis is calculated to be 13.256 mg/L. The invention provides the application of the Osmunda japonica in inhibiting the survival rate of fish melon insects under the condition of no toxicity concentration to cells for the first time, provides a novel medicine for preventing and treating the infection of the fish melon insects of aquaculture animals, is a natural medicinal plant, is environment-friendly, and has the advantages of reducing the risk of medicine residue and the like.

Description

Application of rhizoma Osmundae in preventing and treating ichthyophthiriasis of aquaculture animals Technical Field The invention belongs to the technical field of aquaculture medicines, and particularly relates to application of rhizoma Osmundae in preparation of medicines for preventing and treating ichthyophthiriasis infection. Background Aquaculture has made significant progress in terms of scale and technology in recent years as an important industry for securing global food safety and economic development. However, with the increase of the culture density and the complexity of the culture environment, the parasite problem is increasingly prominent, and becomes one of the key factors restricting the sustainable development of aquaculture. The melon (Ichthyophthirius multifiliis) (referred to as melon) belongs to the genus melon of the phylum Protopanaceae, ciliate, quan Maomu. Its life history is divided into glancing food, trophozoite, capsule precursors and capsules. The predator is driven into the host's epidermis by a "drill" at the tip of the parasite, and then differentiated into trophozoites which feed on the host's tissue fragments, secreted mucus and cells, and slowly rotate in the host's epithelium. The mature trophozoite becomes a precursor of the capsule after being separated from the fish body, can do free movement, then secretes a layer of colloid thick film to form the capsule, and is immersed into the water or attached to aquatic weeds and plant scraps. The insect bodies in the capsule become grazing bodies after multiple splitting, and the grazing bodies freely swim in water and find hosts for parasitism. The grazing food body is the only period with infectivity in the life history of the melon insects, and the period is the key link for preventing and treating the melon insects. The ichthyophthirius is mainly parasitic on the fish body and gills, so that the fish body secretes a large amount of mucus, and the fish is difficult to breathe. Many small white spots appear in the parasitic site where epithelial cells proliferate, so ichthyophthiriasis is also known as ichthyophthiriasis. The ichthyophthirius causes damage to the surface skin of fish, increases the chance of other pathogenic infection, increases the death rate of fish, and causes huge economic loss to the aquaculture and ornamental fish industry. Chemicals (malachite green, mercurous nitrate) are listed as forbidden drugs for high toxicity, teratogenicity and carcinogenesis as effective drugs for controlling ichthyophthiriasis. In recent years, plant extracts have a remarkable effect in killing melon insects. The 5% stock solution of the capsicum water extract can obviously kill 90% grazing food after being diluted by 32 times, and 90% grazing food can be killed after being soaked in 150mg/L of the mucuna pruriens leaf formaldehyde extract and 200mg/L of the papaya seed petroleum ether extract for 6 hours. The plant extract has great research prospect in development of anti-ichthyophthirius medicines due to the characteristics of low toxicity, environment friendliness, high efficiency and the like. Osmunda japonica is a perennial herb plant which mainly grows in warm temperate zones and subtropical areas of the south of Qinling, is common in acid soil, has cold nature and bitter taste, and has the effects of clearing heat, detoxicating and stopping bleeding. It is widely used for treating various diseases such as common cold, dysentery, etc. In addition, osmunda japonica thunb also shows remarkable effects of expelling parasites and resisting viruses. The decoction has a certain inhibition effect on ascariasis of swine in vitro, and also has strong resistance to various viruses such as adenovirus type 3 and the like. Meanwhile, the rhizoma osmundae can also influence the blood coagulation process, shorten the blood coagulation time and have obvious hemostatic effect. In conclusion, the Osmunda japonica Thunb plays an important role in the field of traditional Chinese medicines and daily life by virtue of unique medicinal value and economic value. With the deep research of Osmunda japonica, more pharmacological actions and application values of the Osmunda japonica are expected to be revealed and applied. Disclosure of Invention The invention provides application of Osmunda japonica in resisting the infection of the melon insects in the aquaculture animals, wherein the Osmunda japonica water extract can directly inhibit the survival rate of the grazing bodies of the melon insects, and the death number of the grazing bodies is obviously increased along with the increase of the concentration of the medicine, so that the risk of the infection of the aquaculture animals by the melon insects is reduced. In order to achieve the above purpose, the present invention adopts the following technical scheme: toxicity test of Osmunda japonica Thunb on grass carp kidney Cells (CIK) the maximum nontoxic concentration of Osmunda japonica T
