CN-121987678-A - American ginseng poria cocos medicinal and edible composite traditional Chinese medicine composition and application thereof
Abstract
The invention discloses a medicinal and edible composite traditional Chinese medicine composition of American ginseng and poria cocos and application thereof. The composition is prepared from American ginseng powder, poria cocos powder mixture, potassium hydroxide, L-arginine and L-ascorbic acid. The preparation method comprises the steps of carrying out constant-temperature reflux extraction on composite medicinal material powder by adopting a potassium hydroxide aqueous solution to promote depolymerization and dissolution of polysaccharide, collecting an alkaline extract, adding L-arginine for constant-temperature incubation, utilizing guanidine groups of the alkaline extract to combine with polysaccharide chains to form steric hindrance, dropwise adding L-ascorbic acid to neutralize to a pH value of 6.0-6.5 under the protection of nitrogen and in a high-shear dispersion state, and then concentrating under reduced pressure and spray drying to obtain a finished product. According to the invention, through the arginine incubation and high-shear neutralization processes, polysaccharide precipitation is prevented, ginsenoside degradation caused by local peracid is avoided, and meanwhile, the generated organic salt reduces the risk of moisture absorption and agglomeration of the product, so that the extraction retention rate of active ingredients is effectively improved.

Description
American ginseng poria cocos medicinal and edible composite traditional Chinese medicine composition and application thereof Technical Field The invention relates to the technical field of traditional Chinese medicine pharmacy and health-care food processing, in particular to a medicinal and edible composite traditional Chinese medicine composition of American ginseng and poria cocos and application thereof. Background American ginseng and poria cocos are common medicinal and edible medicinal materials. The main active ingredient of American ginseng is ginsenoside, and the main active ingredient of poria cocos is pachyman. In the existing traditional Chinese medicine extraction process, water extraction is the most common treatment mode. However, because of the compact hydrogen bond network structure among pachyman molecules, the conventional water extraction process has low extraction rate of pachyman, and it is difficult to fully release the active ingredients of medicinal materials. In order to improve the dissolution rate of pachyman, the process of alkali liquor extraction and inorganic acid neutralization is commonly adopted in the industry at present. However, this process has several drawbacks in dealing with the complex extraction system of the American ginseng-poria cocos. After the alkali liquor is utilized to break polysaccharide hydrogen bonds so as to improve the extraction rate, inorganic acid such as hydrochloric acid and the like is generally added for neutralization so as to adjust the pH value of the system. The process can introduce high-concentration inorganic salt ions into an extraction system, so that the extract powder prepared by final drying is extremely easy to absorb moisture and agglomerate, and the physical and chemical stability of the product and the subsequent preparation processing are seriously affected. Meanwhile, when acid liquor is dripped into alkaline extracting solution under the condition of conventional mechanical stirring, fluid around the dripping point is difficult to mix evenly instantaneously, and a local high-acidity microenvironment is easy to form. Ginsenoside in American ginseng and extracted polysaccharide components are sensitive to acidic environment, and the local peracid phenomenon is extremely easy to cause the hydrolysis and degradation of the active components. In addition, along with the decrease of the overall pH value of the system in the neutralization process, the depolymerization and dissolution pachyman molecules are easy to generate protonation under the alkaline condition, and then are re-associated and separated out to generate precipitation, so that the actual effective retention rate of the water-soluble polysaccharide in the final fluid extract or dry powder is greatly reduced. Disclosure of Invention The invention solves the technical problems that the extraction rate of pachyman by the existing water extraction process is low, and the conventional alkali extraction acid precipitation or hydrochloric acid neutralization process can introduce high-concentration inorganic salt to cause moisture absorption and agglomeration of the extract, and the neutralization process is easy to generate local high acidity to cause degradation of ginsenoside and polysaccharide. In order to solve the problems, the invention provides the following technical scheme: in a first aspect, the invention provides a medicinal and edible composite traditional Chinese medicine composition of American ginseng and poria cocos, which adopts the following technical scheme: The American ginseng and poria cocos medicinal and edible composite traditional Chinese medicine composition is prepared from the following raw materials, by weight, 100 parts of composite medicinal material mixed powder, 8.4-22.4 parts of potassium hydroxide, 5.0-8.0 parts of L-arginine and L-ascorbic acid, wherein the addition amount of the L-ascorbic acid is the amount required for adjusting the pH value of an extracting solution to 6.0-6.5, and the composite medicinal material mixed powder consists of American ginseng powder and poria cocos powder. By adopting the technical scheme, the compound medicinal material powder, potassium hydroxide, L-arginine and L-ascorbic acid with specific proportions are adopted, so that the compound Chinese medicinal composition with high polysaccharide extraction rate, low inorganic salt load and high active ingredient retention rate is obtained. The specific principle is that potassium hydroxide is used as alkaline medium to break down the hydrogen bond network among pachyman molecules to depolymerize and dissolve out polysaccharide, L-arginine is combined with depolymerized polysaccharide chain to form steric hindrance under alkaline condition to prevent re-association precipitation of polysaccharide in neutralization stage, L-ascorbic acid is used as organic neutralizer to react with potassium hydroxide in system to generate potassium ascorbate organic salt,
