Description

Traditional Chinese medicine composition for inhibiting oral pathogenic bacteria and preparation and application thereof Field of the art The invention belongs to the technical field of biology, and particularly relates to a traditional Chinese medicine composition for inhibiting oral pathogenic bacteria, and preparation and application thereof. (II) background art Caries and bad breath are common oral diseases in clinical practice, and the prevalence rate is up to more than 50%, and the health and life quality of human bodies are seriously affected. Streptococcus mutans (Streptococcus mutans, S.mutans) acts as a major cariogenic bacterium in the oral cavity by producing acid and resisting acids, synthesizing extracellular polysaccharides, forming biofilms, and the like. Mature plaque biofilms may exhibit greater toxicity, resistance to drugs, and ability to resist host immune defenses relative to planktonic bacteria, thus inhibiting plaque biofilm formation and scavenging mature biofilms are critical for caries prevention and treatment. Biofilms are aggregates of microorganisms encapsulated in a self-produced extracellular polymeric matrix. The formation of a dental biofilm is a complex process in which Streptococcus mutans first adheres to the surface of teeth, and then colonizes and accumulates with other oral bacteria to form dental plaque. During this process, streptococcus mutans synthesizes water-insoluble glucans via glucosyltransferase (GTFase) which provide binding sites for bacteria to adhere to the tooth surface and promote biofilm formation. Thus, inhibition of formation of a biofilm by Streptococcus mutans is one of the main strategies for caries prevention. At present, most of clinic treatments for oral diseases use antibiotics or antifungal drugs, but long-term use of the antibacterial drugs easily causes oral microbial flora disturbance, and the biofilm state can protect microorganisms in the oral microbial flora from being damaged by the antibacterial drugs and a host immune system. Fluoride plays an important role in the prevention of caries, but fluoride at a high concentration is cytotoxic. Chlorhexidine is also a commonly used anticaries agent, however frequent use of chlorhexidine accelerates the formation of dental calculus. Thus, there is a need to find a biofilm replacement therapeutic strategy to combat oral disease and maintain oral health. In recent years, research on caries prevention using natural medicines has been widely focused by scholars at home and abroad, and has been greatly progressed. The traditional Chinese medicine in China has great potential in regulating the micro-ecological balance of the oral cavity and preventing the infectious diseases of the oral cavity as a main natural medicine. Research shows that most of Chinese herbal medicines have better antibacterial effect. Compared with the traditional anticarious medicine, the Chinese herbal medicine has the advantages of high efficiency, no pollution, small toxic effect, low drug resistance and the like, so that the preparation is more suitable for clinical application. However, although there have been several extracts of Chinese medicinal materials that have demonstrated the potential to inhibit Streptococcus mutans in experiments, the effects of natural drugs on mixed biofilms, synergistic effects between natural drugs, and combinations of natural drug anticaries with other anticaries means have yet to be further studied. (III) summary of the invention The invention aims to provide a traditional Chinese medicine composition for inhibiting oral pathogenic bacteria, and a preparation method and application thereof, wherein the traditional Chinese medicine composition comprises extracts of natural medicines of rhizoma anemarrhenae and coptis chinensis, can synergistically inhibit growth of streptococcus mutans and formation of biofilm, provides a new thought for treating oral diseases, promotes application of traditional Chinese medicine extracts in oral care products, and provides an effective alternative scheme for reducing oral diseases and reducing negative effects caused by abuse of antibiotics. The technical scheme adopted by the invention is as follows: the invention provides a traditional Chinese medicine composition for inhibiting oral pathogenic bacteria, which comprises one of an alcohol extract (preferably an ethanol extract) of rhizoma anemarrhenae or stir-fried rhizoma anemarrhenae or an organic solvent extract phase of the alcohol extract, and one of an aqueous extract or an organic solvent extract phase of the aqueous extract of coptis chinensis. Further, the organic solvent is n-butanol or ethyl acetate. Further, the traditional Chinese medicine composition consists of an n-butanol extract phase of an alcohol extract of rhizoma anemarrhenae or stir-fried rhizoma anemarrhenae and an n-butanol extract phase of a water extract of coptis chinensis. Further, the mass ratio of the n-butanol extract ph
