Abstract

The invention discloses a germinated brown rice extract capable of remarkably improving liver cholesterol metabolism, and a preparation method and application thereof. The method comprises soaking the cleaned and sterilized brown rice in inositol water solution, and spraying inositol water solution with the same concentration during germination. And after germination, drying and crushing brown rice, adding a methanol aqueous solution, adding ascorbic acid with a certain concentration, ultrasonically extracting, and centrifuging to obtain a supernatant to obtain a target extract. Compared with the conventional germinated brown rice alcohol extract, the germinated brown rice extract prepared by the invention has better polyphenol enrichment effect and higher polyphenol retention rate in the ultrasonic extraction process, and the prepared extract can obviously regulate liver cholesterol metabolism.

Description

Preparation and application of germinated brown rice extract capable of remarkably improving liver cholesterol metabolism Technical Field The invention relates to the technical field of grain deep processing and functional component extraction, in particular to a germinated brown rice extract capable of remarkably improving liver cholesterol metabolism, and a preparation method and application thereof. Background Under the background of modern dietary structural changes and chronic metabolic diseases, improving liver cholesterol metabolism has become an important strategy for preventing and controlling cardiovascular metabolic diseases such as hyperlipidemia, fatty liver, atherosclerosis and the like. Liver is used as a key organ for synthesizing, metabolizing and regulating cholesterol, and abnormal functions directly affect blood lipid balance. Therefore, the liver cholesterol metabolic process is interfered by natural food-borne components, and the liver cholesterol metabolic process has important health significance and application value. Brown rice is used as common whole grain, is rich in various bioactive substances such as polyphenol, dietary fiber, phytosterol, gamma-aminobutyric acid (GABA) and the like, and researches show that the extract has the potential of regulating lipid metabolism, inhibiting endogenous synthesis of cholesterol and resisting oxidization, wherein the polyphenol component is considered as one of core functional factors for regulating liver cholesterol metabolism. In order to increase the content of active ingredients, germination treatment is often adopted to activate an endogenous enzyme system of brown rice so as to promote the enrichment of ingredients such as polyphenol. However, the existing preparation method of the germinated brown rice active substance still has obvious limitations that, on one hand, an exogenous chemical regulator is often used for promoting germination in the germination process, possibly causing residual risks, and the process is complex and has low controllability, and on the other hand, the subsequent polyphenol extraction is carried out in a mode of combining alcohol extraction with ultrasonic assistance, but the active ingredient is easily damaged by oxidation in the extraction process, so that the yield and the retention rate are reduced, and finally the biological activity of the extract is influenced. Based on the method, the preparation method of the germinated brown rice extract is simple and convenient in process, safe and residue-free, and can synergistically enhance the enrichment and retention of active ingredients, so that the application efficiency of the germinated brown rice extract in improving the cholesterol metabolism of the liver is significant. Disclosure of Invention Aiming at the problems, the invention aims to provide the germinated brown rice extract capable of remarkably improving liver cholesterol metabolism, and the preparation method and application thereof, wherein inositol is selected as a polyphenol enrichment promoter in the germination stage, and simultaneously an antioxidant is added in the extraction stage, so that enrichment, stable protection and terminal nondestructive retention of cholesterol-reducing components such as polyphenol are realized, and the defects in the preparation process of the existing germinated brown rice extract are overcome. In view of the above-mentioned objects, the present invention provides a preparation method of germinated brown rice extract for significantly improving liver cholesterol metabolism, specifically comprising the following steps: s1, washing brown rice with clear water until no impurities exist, soaking, sterilizing and cleaning the brown rice in a sodium hypochlorite solution, and soaking the sterilized brown rice in an inositol aqueous solution; S2, placing the brown rice which is soaked in the step S1 and drained is placed in a culture dish paved with gauze for germination for 24-48 hours, and spraying an inositol aqueous solution in the period; s3, placing the brown rice subjected to germination treatment in the step S2 into a hot air drying oven for drying treatment, and crushing and sieving to obtain germinated brown rice powder; S4, adding ascorbic acid into the methanol solution to obtain a mixed extraction solvent, adding the germinated brown rice powder prepared in the step S3 into the mixed extraction solvent for ultrasonic extraction, centrifuging after ultrasonic extraction is completed, taking supernatant, removing organic solvent by reduced pressure evaporation to obtain a concentrate, and freeze-drying the concentrate to obtain the solid germinated brown rice extract. In one embodiment of the invention, the brown rice is disinfected by using a sodium hypochlorite solution with the mass concentration of 0.1-0.8%, the soaking time is 10-30 min, the soaking temperature is 20-35 ℃, and deionized water is used for washing after the soaking treatment is com
