Abstract
The invention relates to the field of plants, in particular to GhICDH gene and application thereof in regulating and controlling verticillium wilt resistance of plants. The invention provides an application of GhICDH gene or GhICDH protein coded by the GhICDH gene in any of improving verticillium wilt resistance of plants or cultivating new germplasm with high verticillium wilt resistance. According to the invention, through systematically analyzing the sequence characteristics, protein subcellular localization and ubiquitination modification site functions of the cotton GhICDH gene, the disease-resistant mechanism of the gene for regulating and controlling cotton active oxygen burst, callose accumulation and lignin synthesis and the action relation between the gene and the interaction protein GhHSP 70 are clarified, a novel target gene for genetic improvement of cotton verticillium resistance is dug, and the core technical problems that the high-quality disease-resistant gene is deficient, the key regulation mechanism is unknown and high-efficiency molecular breeding is difficult to realize in cotton verticillium resistance research are solved.

Claims (10)
- Use of the ghicdh3 gene or the GhICDH protein encoded by the gene in any of the following: (a) The application of the composition in improving the verticillium resistance of plants; (b) The application in cultivating new germplasm with high verticillium resistance; the nucleotide sequence of GhICDH gene is shown as SEQ ID NO. 1; the amino acid sequence of GhICDH protein is shown as SEQ ID NO. 2.
- 2. The use according to claim 1, wherein the step of increasing verticillium resistance in a plant is carried out by overexpressing GhICDH genes in the plant.
- 3. The method of claim 1 or 2, wherein the plant comprises cotton.
- 4. A method for increasing verticillium resistance in plants, comprising overexpressing the GhICDH gene in plants for use according to any one of claims 1 to 3.
- 5. A method of growing plants with high verticillium resistance, comprising overexpressing the GhICDH gene in plants for use according to any one of claims 1 to 3.
- A ghicdh3 gene mutant characterized by having: (1) An amino acid sequence shown as SEQ ID NO. 3, or (2) An amino acid sequence obtained by substituting, deleting or adding one or more amino groups to the amino acid sequence shown in (1), and functionally identical or similar to the amino acid sequence shown in (1).
- 7. A nucleic acid molecule encoding the GhICDH gene mutant according to claim 6, having: (3) Nucleotide sequence shown as SEQ ID NO. 4, or (4) A nucleotide sequence obtained by modifying, substituting, deleting or adding one or more bases to the nucleotide sequence according to (3), or (5) A sequence having at least 80% homology with the nucleotide sequence of (3) or (4), or (6) The nucleotide sequence of (3), (4) or (5).
- 8. Use of the GhICDH gene mutant according to claim 6 and/or the nucleic acid molecule according to claim 7 in any of the following: (c) The application of the composition in improving the verticillium resistance of plants; (d) The method for improving verticillium wilt resistance of plants comprises the step of over-expressing the nucleic acid molecules in plants, wherein the plants comprise cotton.
- 9. A method for increasing verticillium resistance in plants comprising overexpressing a nucleic acid molecule according to claim 7 in a plant.
- 10. A method for growing plants with high verticillium resistance, comprising overexpressing the nucleic acid molecule of claim 7 in the plant.
Description
GhICDH3 gene and application thereof in regulating and controlling verticillium wilt resistance of plants Technical Field The invention relates to the field of plants, in particular to GhICDH gene and application thereof in regulating and controlling verticillium wilt resistance of plants. Background Cotton is a worldwide important commercial crop, the fibers of which are the core raw material for the textile industry. Verticillium wilt is a soil-borne vascular bundle disease caused by verticillium dahliae, which causes wilting of cotton plants and yellowing and drying of leaves, and causes death of the plants when serious, and becomes a primary biological stress factor for restricting improvement of cotton yield and quality. At present, chemical prevention and treatment means are easy to cause environmental pollution, and are difficult to thoroughly remove pathogenic bacteria in soil, and the traditional disease-resistant breeding has the defects of long period, lack of resistance sources and easy loss of resistance along with the variation of pathogenic bacteria. In molecular mechanism research of plant interaction with pathogenic bacteria, active oxygen outbreaks, callose deposition and lignin synthesis are key defense reactions of plants against pathogenic bacteria infection. Ubiquitination modification is an important protein posttranslational modification mode and can be involved in regulating and controlling the immune response process of plants. The succinic acid signaling pathway is a core pathway for plant energy metabolism and substance synthesis, with the key enzyme isocitrate dehydrogenase (ICDH) playing an important role in regulating carbon metabolism and redox homeostasis. However, the ubiquitination modification characteristics of ICDH family genes in cotton and the specific action mechanism of the ubiquitination modification characteristics in regulating verticillium wilt resistance have not been systematically analyzed. Disclosure of Invention In view of this, the present invention provides GhICDH gene and its use in regulating verticillium resistance of plants. The invention aims at the problems that the existing cotton verticillium resistance research lacks a core disease resistance gene which can be efficiently utilized, the disease resistance of a disease resistant variety cultivated by the traditional breeding means is easy to be lost due to the mutation of pathogenic bacteria, the molecular regulation mechanism of cotton for resisting verticillium infection is not deeply analyzed, particularly the ubiquitination modification of key proteins in a succinic acid signal path and the research of mediated disease resistance paths are insufficient, the sequence characteristics of cotton GhICDH genes, the protein subcellular localization and ubiquitination modification site functions are systematically analyzed, the disease resistance mechanism of the gene for regulating cotton active oxygen burst, callose accumulation and lignin synthesis and the action relation with interaction protein GhHSP 70 are clarified, and a novel target gene for resisting verticillium genetic improvement of cotton is dug, so that the core technical problems of high-quality disease resistance gene deficiency, unknown key regulation mechanism and difficulty in realizing high-efficiency molecular breeding in cotton verticillium resistance research are solved.
