Abstract

The invention discloses an application of InDel molecular marker related to nipple heterosis on a No. 16 chromosome of a pig, wherein the locus of the InDel molecular marker is rs793470896, the InDel molecular marker corresponds to GGAGGCAC > G mutation at 74359589 bp on a No. 16 chromosome of an international pig reference genome 11.1 version, and the genotype of the InDel molecular marker is GGAGGCAC/GGAGGCAC, GGAGGCAC/G or G/G. The InDel molecular marker provided by the invention is used for marker-assisted selective breeding, pigs with genotype GGAGGCAC/GGAGGCAC in a group are gradually eliminated, and the nipple number of a bred sow can be remarkably increased, so that the breeding and lactation capacity of the sow can be remarkably improved, the survival rate of piglets can be increased, and the production income can be increased.

Claims (8)

  1. 1. The application of the product for detecting the InDel molecular marker related to nipple heterosis on the chromosome 16 of the pig is characterized in that the locus of the InDel molecular marker is rs793470896, and the application comprises at least one of the following (1) - (4): (1) Identifying the nipple number heterosis of the pigs; (2) Preparing a product for identifying the nipple number heterosis of pigs; (3) Pig genetic improvement based on elimination of the InDel molecular marker genotype GGAGGCAC/GGAGGCAC pigs to improve nipple number heterosis of the pigs; (4) A product is prepared for assisting in genetic improvement of pigs, which product enables assisting in genetic improvement of pigs based on identification of the genotype of the InDel molecular marker.
  2. 2. The use according to claim 1, wherein the product for detecting the InDel molecular markers associated with Nipple heterosis on chromosome 16 of pigs comprises at least one of reagents, kits, chips and devices for detecting the InDel molecular markers.
  3. 3. The use according to claim 2, wherein the reagent for detecting the InDel molecular marker comprises at least one of a primer, a probe for detecting the InDel molecular marker.

The use according to claim 3, wherein the primer for detecting the InDel molecular marker comprises an upstream primer and a downstream primer, the nucleotide sequence of the upstream primer is shown as SEQ ID NO. 2, and the nucleotide sequence of the downstream primer is shown as SEQ ID NO. 3.
  5. 5. The use according to any one of claims 1 to 4, wherein the pig is a ternary hybrid pig.

A method for genetic modification of pigs comprising the steps of: (1) Determining the genotype of InDel molecular markers related to papillary heterosis on chromosome 16 of the pig; (2) Individuals with InDel molecular marker genotype GGAGGCAC/GGAGGCAC are eliminated; the locus of the InDel molecular marker is rs793470896.

The method of claim 6, wherein in step (1), the method of determining the genotype of the InDel molecular marker associated with Nipple heterosis on chromosome 16 of swine comprises the steps of: Extracting pig whole genome DNA, carrying out PCR amplification by using a primer pair with nucleotide sequences shown as SEQ ID NO. 2 and SEQ ID NO. 3, sequencing the amplified product, and determining the genotype of InDel molecular marker related to nipple number heterosis on the chromosome 16 of the pig based on the sequencing result.

Application of InDel molecular marker on pig chromosome 16 and related to nipple number heterosis Technical Field The invention belongs to the technical field of molecular biology, and relates to application of InDel molecular markers on a pig chromosome 16 and related to nipple heterosis. Background The nipple number (Total Teat Number, TTN) of the sow is taken as one of reproductive traits, is an important index for measuring the lactation and lactation capacity of the sow, has a profound effect on the reproductive performance of the sow, and is an important index for evaluating the reproductive capacity of the breeding sow in production. The number of the sow nipple is closely related to the survival rate and the growth and development condition of piglets, in the whole industry chain of live pigs, the number of the sow nipple and the litter size are closely related, and more piglets can be fed to sows with more nipple numbers, so that the survival rate and the weaning litter weight of the piglets are improved, and the economic benefit of a pig farm is further increased. The hybrid vigor (Heterosis) refers to the phenomenon that the properties of the hybrid offspring of different varieties or strains of species in growth, development, propagation, disease resistance, stress resistance and the like are superior to those of the parent, and the excellent properties of different varieties can be combined through hybridization breeding, so that the ideal production properties are obtained. In recent years, duchesner long ternary hybrid pigs (Duroc x long white pigs x large white pigs) occupy most commercial pig markets, and the hybrid combination is excellent in growth speed, feed utilization efficiency, lactation capacity and the like through long-term manual selection. Disclosure of Invention The invention aims to provide an application of InDel molecular markers which are located on a pig chromosome 16 and are obviously related to nipple trait heterosis. According to one aspect of the invention, there is provided the use of a product for detecting InDel molecular markers associated with Nipple heterosis on chromosome 16 of pigs, the use comprising at least one of the following items (1) - (4): (1) Identifying the nipple number heterosis of the pigs; (2) Preparing a product for identifying the nipple number heterosis of pigs; (3) Pig genetic improvement is based on elimination of the pig with InDel molecular marker genotype GGAGGCAC/GGAGGCAC so as to improve nipple number heterosis of the pig; (4) A product for assisting in genetic improvement of pigs is prepared, and the product realizes assisting in genetic improvement of pigs based on identification of genotypes of InDel molecular markers. The InDel molecular marker which is positioned on the chromosome 16 of the pig and is obviously related to the nipple number heterosis provided by the invention, the locus is rs793470896, the deletion mutation corresponding to 7 nucleotides (GAGGCAC) on the chromosome 11.1 version 16 of the international pig reference genome is positioned between 74359590 bp and 74359596 bp, or the mutation is called GGAGGCAC > G mutation corresponding to 74359589 bp on the chromosome 11.1 version 16 of the international pig reference genome, and the difference of the parental dominance in the nipple number trait of the pig is caused. InDel molecular markers have genotypes GGAGGCAC/GGAGGCAC (wild type), GGAGGCAC/G (heterozygous deleted) or G/G (homozygous deleted). The InDel molecular marker provided by the invention is obviously related to the nipple number trait heterosis of pigs, wherein the total nipple number of the pigs with the gene type of GGAGGCAC/G of the InDel molecular marker is obviously larger than that of the pigs with the gene type of GGAGGCAC/GGAGGCAC (P < 0.001), and the InDel mutation is favorable for the nipple number heterosis of the pigs. The identification of the nipple number heterosis of the pig can be realized by detecting whether the pig only carries the InDel molecular marker, and in the hybridization breeding of the pig, the breeding process of the pig can be accelerated and the reproductive capacity of the sow can be improved by breeding the pig with the InDel molecular marker genotype GGAGGCAC/G or G/G.
