Abstract

The invention discloses an application of RsBLV virus in preparation of a biological control preparation of tobacco target spot bacteria, and belongs to the technical field of tobacco target spot bacteria control. The invention extracts a new Benyviridae virus RsBLV, rsBLV virus RNA sequence shown in SEQ ID NO.3 from tobacco target spot germ GZ-6 carrying single virus. The pathogenicity of the original strain GZ-6 and the detoxified tobacco target spot germ GZ-6 is compared by an in-vitro leaf method, and the statistical plaque size shows that the lesions caused by the strains infected by RsBLV are obviously smaller than those of homologous virus-removing strains, so that the virus RsBLV can reduce the pathogenicity of fungi, and the method can be applied to preparing biocontrol agents for inhibiting the pathogenicity of the tobacco target spot germ.

  1. The application of RsBLV8 virus in preparing biological control preparation of tobacco target spot bacteria is characterized in that RsBLV virus RNA sequence is shown as SEQ ID NO. 3; the biocontrol agent is a biocontrol agent for inhibiting the pathogenicity of tobacco target spot bacteria.
  2. 2. A biocontrol formulation comprising the RsBLV virus of claim 1, wherein the biocontrol formulation comprises RsBLV virus particles and an agriculturally acceptable carrier.

Application of RsBLV virus in preparation of biocontrol agent of tobacco target spot germ Technical Field The invention relates to the technical field of prevention and control of tobacco target spot bacteria, in particular to application of RsBLV virus in preparation of a biological prevention and control preparation of tobacco target spot bacteria. Background Rhizoctonia solani (Rhizoctonia solaniK ulhn) is a asexual generation of the basidiomycete leather melon (Thanatephorus cucumeris (Frank) Donk), belonging to the genus Rhizoctonia. Rhizoctonia solani is soil-borne plant pathogenic bacteria with wide parasitic range, infects crops such as tobacco, potato, rice, soybean, corn, wheat and the like, is extremely easy to induce diseases such as seedling rot, damping off, root rot and the like, and endangers the planting yield of the crops. Wherein, the potato black nevus bacteria and the tobacco target spot bacteria which are the rhizoctonia solani AG-3 fusion group respectively cause serious stem basal diseases and leaf diseases to potatoes and tobacco. With the increasing maturation of metagenome and transcriptome technologies, a large number of mycoviruses existing in Rhizoctonia solani are well known, so that viruses related to the pathogenic decline of Rhizoctonia solani are screened and identified, and screening of mycoviruses with attenuated properties through the diversity of related mycoviruses and their interaction relationship with hosts is also a research hotspot in the related field. Disclosure of Invention The invention aims to provide an application of RsBLV virus in preparation of a biological control preparation of tobacco target spot bacteria, and the fungus virus and the characteristic of being capable of weakening pathogenicity of rhizoctonia solani are utilized to lighten the harm of the tobacco target spot bacteria to crops, so that biological control of the rhizoctonia solani is realized, and theoretical basis and guidance are provided for developing a novel biological control bacterial agent. In order to achieve the above purpose, in one aspect, the invention provides an application of RsBLV virus in preparing a biocontrol agent for inhibiting pathogenic force of tobacco target spot bacteria, wherein the RNA sequence of RsBLV virus is shown as SEQ ID NO. 3. RsBLV8 after a host is infected, key regulation nodes such as a glycoside hydrolase family, glutathione S-transferase, zinc cluster transcription factors, bZIP family, CFEM effector protein, MFS superfamily and the like are obviously influenced, and the genes remodel a physiological metabolism network of fungi by regulating carbon source metabolism, oxidative stress response, secondary metabolite synthesis and host immunosuppression ways, so that hypha growth, stress tolerance and pathogenic factor secretion are finally influenced, and the pathogenicity of the strain is enhanced or weakened. (3) RsBLV8 is a novel virus which is not reported in Benyviridae families, the separation and identification of the novel virus expand the diversity of fungal viruses, and a novel model is provided for the research of virus-host interaction. (4) RsBLV8 shows remarkable biocontrol potential by inhibiting host pathogenicity, slowing down hypha growth and regulating key metabolic pathways.
