CN-122004483-A - Composite prebiotic composition for regulating kidney-intestine axis and delaying aging and application thereof
Abstract
The invention belongs to the technical field of functional foods, health-care products and medicines, and provides a composite prebiotic composition for regulating kidney-intestine axis and delaying aging and application thereof. The composite prebiotic composition contains 7.5 parts of fructo-oligosaccharide, 0.035 parts of yeast beta-glucan, 17.5 parts of resistant starch and 0.12 parts of luteolin. The formula combines fermentable carbohydrate with immunoregulatory polysaccharide and antioxidant polyphenol, and adjusts intestinal microecology and systemic state by providing multi-target synergistic effects of multiple carbon sources, regulating immunity, resisting inflammation and oxidation and the like. Animal experiments show that the Chinese medicinal composition can interfere with kidney-intestine axis dysfunction, can delay skin and hair aging after being fed to an aged mouse for a long time, has long health life and good safety, and is suitable for developing related products of intestinal health, kidney-intestine axis diseases and aging resistance.

Description
Composite prebiotic composition for regulating kidney-intestine axis and delaying aging and application thereof Technical Field The invention belongs to the technical field of functional foods, health-care products and medicines, and particularly relates to a composite prebiotic composition for regulating kidney-intestine axis and delaying aging and application thereof. Background The intestinal flora interacts intimately with distant organs (e.g. kidneys) via the "kidney-gut axis". In Chronic Kidney Disease (CKD) and other conditions, dysbacteriosis in the intestinal tract can lead to increased production of uremic toxins (e.g., indoxyl sulfate, p-cresol), exacerbating systemic inflammation and organ damage. At the same time, the aging process itself is accompanied by a decrease in intestinal flora diversity and a decrease in beneficial bacteria. The prebiotics are effective means for regulating intestinal flora as "nutrients" for the beneficial bacteria. Most of the current prebiotics are single components (such as inulin and fructo-oligosaccharides) or simple combinations, the effect of which is limited, and the lack of systematic and multi-target designs for complex physiological networks (such as kidney-gut axis and aging, which are involved in metabolism, immunity and oxidative stress). It is difficult in the prior art to clearly obtain evidence of an anti-aging phenotype in long-term safety observations while effectively improving specific pathological models (e.g., renal-intestinal dysfunction). Therefore, a composite prebiotic composition which has definite components and scientific proportion and can act synergistically through a plurality of mechanisms is developed to meet the clinical and health requirements of accurately and comprehensively regulating intestinal microecology, and has important significance. Disclosure of Invention The invention provides a composite prebiotic composition for regulating kidney-intestine axis and deferring senility and application thereof, which is prepared by multi-target synergistic effect, aims to regulate intestinal microecology, improve kidney-intestinal axis dysfunction caused by intestinal flora disturbance, and show physiological benefits of deferring aging in long-term application. The technical scheme of the invention is as follows: in a first aspect of the present invention, a composite prebiotic composition is provided, the active ingredients of which comprise the following components: The functional oligosaccharide comprises, by mass, 5-10 parts of functional oligosaccharide, 0.03-0.04 part of polysaccharide, 15-20 parts of dietary fiber and 0.10-0.14 part of polyphenol. The composite prebiotic composition, wherein the functional oligosaccharide is selected from any one of fructo-oligosaccharide, xylo-oligosaccharide and galacto-oligosaccharide, and preferably the functional oligosaccharide is fructo-oligosaccharide; the polysaccharide is selected from any one of yeast beta-glucan, mushroom polysaccharide and ganoderma lucidum polysaccharide, and preferably, the polysaccharide is yeast beta-glucan; The dietary fiber is selected from any one of resistant starch, pectin and algin, preferably, the dietary fiber is resistant starch, and further preferably, the dietary fiber is RS2 type resistant starch; the polyphenol is selected from any one of luteolin, catechin and anthocyanin, preferably, the polyphenol is luteolin. The above composition may be prepared in any conventional oral dosage form including, but not limited to, capsules, tablets, powders, solid beverages or special medical use formulas. In preparation, the content of each active ingredient should be converted according to the recommended daily intake total and the specifications of unit dosage form. Preferably, the daily intake mass ratio of the four active ingredients in the composite prebiotic composition is 7.5 parts of fructo-oligosaccharide, 0.035 parts of yeast beta-glucan, 17.5 parts of resistant starch and 0.12 parts of luteolin. Further preferably, the four active ingredients in the composite prebiotic composition are prepared from 7.5 parts of fructo-oligosaccharide, 0.035 parts of yeast beta-glucan, 17.5 parts of RS2 resistant starch and 0.12 parts of luteolin by daily intake. In a second aspect, the invention provides the use of the complex prebiotic composition in the manufacture of a medicament, nutraceutical or formulation for the treatment and/or amelioration of diseases associated with dysfunction of the kidney-gut axis. Among these, diseases associated with dysfunction of the kidney-gut axis include chronic kidney disease and its concomitant intestinal flora disorders, constipation or systemic micro-inflammatory states. In a third aspect, the present invention provides the use of a complex prebiotic composition in the manufacture of a health care and/or functional food for the delay of skin aging, maintenance of hair health or extension of healthy life in a mammal.
