Abstract
The invention relates to the field of prevention and control of avian viral diseases, in particular to application of an epimedium extract in preparing a medicament for preventing and/or treating infectious bronchitis virus infection. The herba Epimedii extract is herba Epimedii ethanol extract. The biological safety and antiviral activity of the extract are evaluated by constructing a chick embryo infection model, the epimedium extract has no obvious toxic effect on chick embryo, and the survival rate of chick embryo can be improved by treatment after the chick infectious bronchitis virus infection. Further combined with tissue immunofluorescence detection, western immunoblotting analysis and probe method fluorescence quantitative PCR detection results show that the extract provided by the invention can reduce the expression level of virus antigen and the copy number of virus nucleic acid in chicken embryo liver tissue and allantoic fluid, thereby playing a role in preventing or treating IBV infection. The epimedium extract has the advantages of stable raw material source, simple and convenient preparation process, lower production cost and good popularization and application prospect.

- 1. The application of the epimedium extract in preparing the medicine for preventing and/or treating the infectious bronchitis virus infection of chicken is disclosed, wherein the epimedium extract is an epimedium ethanol extract.
- 2. The use according to claim 1, wherein the use of the epimedium extract for the preparation of a medicament for increasing survival of infected chick embryos, decreasing expression levels of viral antigens in the liver, decreasing copy number of viral nucleic acids in the liver and allantoic fluid.
- 3. The use according to claim 1, wherein the epimedium ethanol extract is prepared by extracting, filtering and concentrating epimedium serving as a raw material and ethanol serving as an extraction solvent, wherein the ethanol is an ethanol aqueous solution with a volume fraction of 60% -95%.
- 4. The use according to claim 3, wherein the extraction is one or more of ultrasonic extraction, reflux extraction or leaching.
- 5. The use according to any one of claims 1 to 4, wherein the epimedium extract and pharmaceutically acceptable excipients are used for preparing a pharmaceutical preparation for resisting infectious bronchitis virus.
- 6. The use according to claim 5, wherein the preparation is one of an oral liquid, a granule, an injection or a spray.

The use according to claim 1, wherein the method for preparing the ethanol extract of epimedium comprises the steps of: (1) Drying and crushing epimedium, and sieving with a 40-80 mesh sieve; (2) Adding the epimedium powder into an ethanol solution with the volume fraction of 60-95% according to the solid-to-liquid ratio of 1:8-20, fully and uniformly mixing, and carrying out ultrasonic extraction for 3 times; (3) After the extraction is finished, centrifuging the extracting solution, taking a supernatant, filtering by medium-speed filter paper, placing the filtrate in a rotary evaporator, recovering ethanol and concentrating to be thick under the conditions of the temperature of 40-50 ℃ and the vacuum degree of minus 0.085 to minus 0.095 MPa and the rotating speed of 80-120 rpm; (4) Vacuum drying the concentrate to constant weight, and pulverizing to obtain ethanol extract of herba Epimedii.

The virus mainly invades the respiratory tract system of chickens and can simultaneously affect a plurality of tissues and organs such as kidneys, reproductive systems and the like. Clinically, the problems of abnormal respiration, sneeze, cough, reduced feed intake, limited growth and development and the like are often manifested, and the problems of reduced laying rate, reduced eggshell quality and the like are often accompanied after the infection of the laying hens. If the control measures are improper, higher morbidity and mortality are easy to cause, and obvious economic loss is caused for poultry production. At present, the prevention and control of IB mainly depend on vaccine immunity and enhanced biological safety management. Herba Epimedii (Epimedium spp.) is used as traditional medicinal plant, and its extract contains icariin, flavonoids, and phenols. At present, systematic research and public report about the application of the epimedium extract in preventing and treating infectious bronchitis viruses of chickens are not yet available, and natural source antiviral candidates for IBV are still limited. Disclosure of Invention Aiming at the problems of rapid IBV transmission, frequent strain variation, limited vaccine cross protection capability, lack of special antiviral treatment means and the like in the prior art, the invention provides an anti-IBV technology derived from natural plants.
