CN-122005652-A - Preparation method of Mongolian medicine patch
Abstract
The invention belongs to the technical field of traditional Chinese medicine preparations, and discloses a preparation method of a Mongolian medicine patch. The patch is a multilayer skeleton type transdermal drug delivery system, sequentially comprises a back lining layer, a drug matrix layer and a protective layer from outside to inside, wherein the drug matrix layer is a double-layer composite hydrogel consisting of a drug storage layer and a controlled release layer, and is prepared by taking mango seeds, hainan rose peaches and Caesalpinia crista as main drug components, extracting with water, precipitating with ethanol, freeze-drying to obtain freeze-dried powder, mixing with a matrix containing a transdermal enhancer, sequentially coating, pre-curing and the like. The patch can soften skin horny layer by adding chemical permeation promoter into matrix, and promote permeation of patch medicine into skin, thereby effectively avoiding the problem of low medicine absorption efficiency.

Description
Preparation method of Mongolian medicine patch Technical Field The invention belongs to the technical field of traditional Chinese medicine preparations, and particularly relates to a preparation method of a Mongolian medicine patch. Background The Mongolian medicine patch is an important form of a Mongolian medicine transdermal administration preparation, is a modern transdermal administration system developed on the basis of traditional Mongolian medicine external preparations such as black plaster and rubber paste, and generally consists of a backing layer, a medicine storage layer, an adhesive layer and an anti-adhesive layer. With the wide application of Mongolian external treatment and the rapid development of polymer chemistry, the research of modern traditional Chinese medicine patches is gradually increased, such as tripterygium wilfordii patch, compound red sage root patch, naoshuantong paste, shuxin paste and the like. At present, traditional Chinese medicine patches can be mainly divided into the following technical types: reservoir type patches, unitary patches, and hydrogel patches. However, the prior art has the following problems: the traditional patch has low drug release and absorption efficiency, wherein the drug active ingredients are usually mixed in a matrix, so that the drug administration concentration is low, the bioavailability is low, and particularly, when the drug active ingredients are only ground to the fineness of a 100-mesh sieve, the bioavailability is more limited; insufficient process stability, improper control of coating process parameters (such as temperature and speed) can easily cause uneven thickness or bubbles in the matrix, and the consistency of drug release is affected. Disclosure of Invention This section is intended to outline some aspects of embodiments of the application and to briefly introduce some preferred embodiments. Some simplifications or omissions may be made in this section as well as in the description of the application and in the title of the application, which may not be used to limit the scope of the application. The present invention has been made in view of the above and/or problems occurring in the prior methods for preparing Mongolian patches. Therefore, the invention aims to provide a preparation method of the Mongolian medicine patch, which is used for transdermal drug delivery, bypasses a digestive system, improves the utilization rate, quickens the effect, reduces the side effects of intestines and stomach, has simple preparation process and convenient use, can reduce the comprehensive cost, provides the Mongolian medicine patch which is safe, effective and good in compliance, is attached to the Shenque acupoint of a human body, can be used for preventing renal aging, further maintains sufficient essence and plays a role in delaying aging. In order to solve the technical problems, according to one aspect of the present invention, the following technical solutions are provided: a preparation method of the Mongolian medicine patch comprises the following steps of pretreatment of medicine raw materials, matrix preparation, medicine mixing, coating and forming, wherein the medicine raw materials comprise mango seeds, hainan rose peaches and large-leaf taro, a chemical permeation promoter is added into a hydrophilic matrix in the matrix preparation step, and a traditional Chinese medicine extract is added into the matrix for multiple times in a small amount and mechanical stirring time is prolonged in the medicine mixing step. As a preferable scheme of the preparation method of the Mongolian medicine patch, the medicine raw materials are mixed and used according to the mass ratio of 1:1:1. The preparation method of the Mongolian medicine patch is characterized by comprising the following steps of extracting the mixed medicine raw materials by adopting a water extraction and alcohol precipitation method, and freeze-drying to obtain the kidney-tonifying three-jelly dry powder. As a preferable scheme of the preparation method of the Mongolian medicine patch, the patch is of a multi-layer framework structure, and comprises a back lining layer, a medicine matrix layer and a protective layer in sequence from outside to inside, wherein the medicine matrix layer is a double-layer composite hydrogel consisting of a medicine storage layer and a controlled release layer.
