In particular, the present invention relates to SMARCA2 inhibitors having the structure of formula (I) and pharmaceutical compositions thereof, their use as SMARCA2 inhibitors and their use in the manufacture of a medicament for the treatment of cancer or tumour associated at least in part with SMARCA 2. Wherein each substituent of formula (I) is as defined in the specification.

Description
Polycyclic SMARCA2 inhibitor and application thereof Technical Field The invention belongs to the field of drug synthesis, and particularly relates to a polycyclic SMARCA2 inhibitor and application thereof. Background The SWI/SNF complex is an ATP dependent chromatin remodeling complex and plays an important role in physiological activities such as DNA repair, replication and transcriptional activation of genes. The complex is composed of multiple subunits, including ATP-dependent catalytic subunits (e.g., smarca2\4), core regulatory subunits (e.g., SMARCB 1), and other variable subunits (e.g., ARID 1A). Studies have reported that the SWI/SNF complex subunit plays an important role in tumor suppression, with about 20% of human malignancies having mutations in the complex subunit. SMARCA4, also known as BRG1, can accomplish activation or inhibition of transcription in the complex by the action of atpase. SMARCA4 is also the most commonly mutated atpase subunit in cancer, and has mutations in common malignant tumors such as lung cancer, colon cancer, bladder urothelial cancer, invasive breast ductal cancer, and the like. Wherein about 10% of non-small cell lung cancer patients carry SMARCA4 mutations. It is reported that patients with non-small cell lung cancer with SMARCA4 homozygous deletion are more likely to develop drug resistance and have poor curative effects. SMARCA2 and SMARCA4 are identical to ATPase subunits of SWI/SNF complex, and they have high homology (> 75%). It has been reported that in SMARCA4 mutated tumor cells, lowering SMARCA2 gene expression by RNAi technology can inhibit proliferation of tumor cells, demonstrating the synthetic lethal relationship of SMARCA2 and SMARCA 4. SMARCA2 knockout mice can grow normally, suggesting safety for targeting SMARCA 2. Patients with SMARCA4 deficiency may therefore have better clinical efficacy with SMARCA2 targeted treatment regimens. There is no approved drug for SMARCA2 on the market, and the need for drug development for this target is urgent. Protein targeting chimera (PROTAC) technology has been attracting attention since the advent of 2001, and a number of drugs based on this technology are now entering the clinical research stage, with some compounds already entering clinical stage 3. PROTAC is used as a heterobifunctional molecule, one end of which is a small molecule ligand compound capable of recognizing target protein, and the other end is a ligand capable of recognizing E3 ubiquitin ligase, and the two are connected through a connecting bond (Linker). PROTAC molecules initiate ubiquitination of target proteins in vivo by forming a ternary complex with the target proteins and E3 ubiquitin ligase, thereby mediating degradation of the target proteins by proteasomes. Therefore, the development of novel SMARCA2 small molecule ligand compounds is of great importance, and PROTAC molecules targeting SMARCA2 can be obtained based on the novel SMARCA2 small molecule ligand compounds.
