The separation method can not only efficiently extract IgG, but also can ensure that the high activity of lactoferrin and alkaline protein is reserved. Meanwhile, the technology can prepare the bovine coloctrum composite protein product rich in various high-activity proteins.

Claims (9)
- 1. A method for separating bioactive proteins in bovine colostrum, comprising: s1, pretreating raw milk to obtain sterilized bovine colostrum; S2, performing first ultrafiltration treatment on the sterilized bovine coloctrum so as to obtain a first permeate and a first trapped fluid containing IgG; s3, performing second ultrafiltration treatment on the first permeate so as to obtain a second permeate and a second trapped fluid containing lactoferrin; and S4, performing third ultrafiltration treatment on the second permeate so as to obtain a third trapped fluid containing alkaline proteins.
- 2. The separation process of claim 1, wherein the first ultrafiltration treatment is performed by filtration and concentration using an ultrafiltration membrane having a molecular weight cut-off of 100 to 120 kDa.
- 3. The separation process of claim 1, wherein the second ultrafiltration treatment is performed by filtration and concentration using an ultrafiltration membrane having a molecular weight cut-off of 30 to 50 kDa.
- 4. The separation method according to claim 1, wherein the third ultrafiltration treatment is performed by filtration and concentration using an ultrafiltration membrane having a molecular weight cut-off of 5 to 10 kDa.
- 5. The separation method according to claim 1, wherein in step S1, the sterilization treatment comprises cross-flow microfiltration at 5-10 ℃ using a microfiltration membrane; Optionally, the microfiltration membrane has a pore size of 0.2-0.45 μm.

Background Bovine colostrum is rich in a variety of bioactive proteins, of which immunoglobulins (especially IgG), lactoferrin and basic proteins are of paramount importance. It has unique values of enhancing immunity, resisting bacteria and viruses, promoting growth, regulating physiological functions, etc. At present, the process for separating specific proteins from bovine coloctrum mainly comprises the following steps: Single IgG extraction process, common methods such as salting out, ultrafiltration, ion exchange chromatography, etc. These methods are generally aimed at achieving high purity extraction of IgG, but other active proteins are often ignored or damaged during the process. For example, severe pH changes or high salt environments can lead to irreversible denaturation of lactoferrin and alkaline proteins. The process of mixing active protein powder, typically by low temperature pasteurization and spray drying, aims to "retain" all active ingredients. Therefore, an object of the present invention is to provide a method for separating bioactive proteins from bovine coloctrum and a method for preparing a composite bovine coloctrum product, which can not only efficiently extract IgG, but also retain high activity of lactoferrin and alkaline proteins. Meanwhile, the invention provides a multi-active protein co-enrichment process which is simple and convenient to operate, mild in condition and suitable for large-scale production.
