Description
Banana streak virus vaccine and application thereof in preventing and treating banana streak virus disease Technical Field The invention belongs to the technical field of bioengineering. More particularly, it relates to banana streak virus vaccines and their use in the prevention and treatment of banana streak virus diseases. Background Banana streak virus (banana streak virus, BSV) belongs to the genus of baculo DNA virus of the family Cauliflower mosaic virus, is a main pathogen causing banana streak virus diseases, is widely distributed in each large banana producing area in China, and typical infection symptoms of the banana streak virus are marked by fading stripes, mottle or necrotic spots on leaves, and plant dwarfing, slow growth, cluster malformation and even sterilization are caused when the infection is serious, so that the yield and commodity value are obviously reduced, and the banana streak virus is one of important pathogens restricting sustainable development of banana industry. BSV has different propagation ways, namely long-distance propagation through toxic asexual propagation materials (such as bud sucking and tissue culture seedling) and field diffusion through mealybugs as mediators. Further complicating matters, BSVs also possess unique "endogenous" characteristics, and a portion of the viral sequences can integrate into the banana host genome to form endogenous banana streak virus (endogenous Banana streak virus, eBSV). Under certain conditions of environmental stress (e.g., high temperature, drought) or in vitro tissue culture, these silent viral sequences may be activated, recombined and released as infectious whole virus particles. The mechanism not only makes the BSV difficult to eradicate, but also greatly increases the difficulty of seedling health detection. At present, no effective prevention and control means exists for BSV, and the prevention and control strategy still mainly depends on source control, including strict screening and cultivation of virus-free seedlings, establishment of standardized detoxification and detection processes, enhancement of field monitoring and timely isolation or eradication of definite disease plants. Therefore, the search for new means for controlling BSV is a major challenge in this field. Banana endogenous viral sequences (banana endogenous virus sequence, BEVs) have been found in banana genomes in recent years (GEERINGET al., 2005), these BEVs contain partial reverse transcriptase (REVERSE TRANSCRIPTASE, RT) and ribonuclease H (RNase H) gene nucleotide sequences, are widely distributed in banana a genome (Musa acuminata) and banana B genome (m. balbisiana), and have become banana genetic material (Bhatet al., 2016). Among the BEVs currently known are BEV UD, BEV UH, BEV NGA, BEV UF, BEV P, BEV Q, BEV UC, BEV UG, etc., among which BEVs BEV UC is most widely distributed in the banana genome. Recently, studies have found a new BEV GZ5 (Chenet al, 2024) containing the complete RT/RNase H in the genome of plantain et al. Although there are a large number of BEVs in the banana genome, the role of these BEVs in BSV-infected bananas is currently unclear. Disclosure of Invention Aiming at the problem that no effective prevention and control means are available for banana streak viruses in the prior art, the invention aims at exploring the relation between Banana Endogenous Viral Sequences (BEVs) and banana streak viruses on banana infection, develops banana streak virus vaccines based on the BEVs, and provides a new efficient prevention and control strategy for banana streak viruses. A first object of the present invention is to provide the use of the endogenous banana streak virus sequences BEV UC and/or BEV GZ5 in the preparation of a product for controlling banana streak virus or preventing banana streak virus diseases. A second object of the present invention is to provide a recombinant plasmid. The third object of the present invention is to provide a recombinant engineering bacterium. The fourth object of the invention is to provide the application of the recombinant plasmid or the recombinant engineering bacteria in preparing products for preventing and controlling banana streak virus or preventing and controlling banana streak virus diseases. A fifth object of the invention is to provide a banana streak virus vaccine. A sixth object of the present invention is to provide a method for controlling banana streak virus or preventing banana streak virus disease. The seventh object of the invention is to provide a preparation method of the recombinant engineering bacteria. The above object of the present invention is achieved by the following technical scheme: The invention discloses application of an endogenous banana streak virus sequence BEV UC and/or BEV GZ5 in preparation of a product for preventing and controlling banana streak virus or preventing banana streak virus diseases, wherein the BEV UC has a sequence shown as SEQ ID NO. 1, and the
