Description
Vibrio paravaliensis phage vB_ VcaS _R45Z and application thereof Technical Field The invention belongs to the field of microorganisms, and particularly relates to a vibrio candidum phage vB_ VcaS _R45Z and application thereof. Background In recent years, with the continuous progress of aquaculture technology, the production mode of aquatic products in China is changed from traditional 'fishing mainly' to 'cultivation mainly'. The novel facilities such as deep-open sea aquaculture, land-based industrial circulating water systems (Recirculating Aquacultural Systems, RAS) and fish and vegetable symbiosis are rapidly developed, so that important support is provided for the large-scale and green production of aquaculture, and the aquaculture yield is promoted to be continuously increased. However, the high-density and intensive cultivation mode aggravates water eutrophication and microecological imbalance, and provides favorable conditions for the propagation of pathogenic microorganisms. Rapid proliferation of pathogens is a significant cause of outbreaks of aquaculture diseases. In aquaculture systems, vibrio (Vibrio) is one of the most common and most serious pathogenic microorganisms. The bacteria are widely distributed in the sea, estuary and salty fresh water environments, and are easy to reproduce especially in warm and eutrophic water bodies. To date, more than 100 species of Vibrio have been identified, many of which are closely related to aquatic animal diseases, including Vibrio parahaemolyticus (V. Parahaemicus), vibrio alginolyticus (V. Alginolyticus), vibrio vulnificus (V. Vulnificus), vibrio canbeijensis (V. Campbellii), vibrio harveyi (V. Harveyi), vibrio canbeijensis (V. Owensii), and the like. The pathogenic vibrio can be rapidly transmitted through water, feed, biological film or co-cultured animals, and various diseases such as septicemia, ulceration, enteritis, luminescent diseases, acute hepatopancreatic necrosis and the like are caused, so that the health of prawns, crabs, shellfish and fishes is seriously threatened. Disclosure of Invention The invention aims to provide vibrio paradise phage vB_ VcaS _R45Z and application thereof. A first object of the present invention is to provide Vibrio candidum phage Vibrio campbellii phage vB _ VcaS _R45Z which was deposited at the microorganism strain collection (GDMCC) of Guangdong province at 8 th year 2025, address: university of road 100 in Vietnam, guangzhou, martyr, post code: 510070, accession number: GDMCC NO. 67441-B1. A second object of the present invention is to provide a microbial preparation for controlling Vibrio candidum or Vibrio anguillarum, which contains the above-mentioned phage Vibrio campbellii phage vB _ VcaS _R45Z as an active ingredient. A third object of the present invention is to provide the use of the above phage Vibrio campbellii phage vB _ VcaS _R45Z or microbial preparation for preventing and/or killing Vibrio candidum or Vibrio anguillarum. The fourth object of the present invention is to provide the use of the phage Vibrio campbellii phage vB _ VcaS _R45Z or microbial preparation described above for the preparation of a product for preventing and/or killing Vibrio paradise or Vibrio anguillarum. Preferably, the Vibrio candidum is Vibrio candidum JL2918. Preferably, the Vibrio anguillarum is Vibrio anguillarum LMG 4437 T Preferably, the product comprises a drug, a disinfectant, a water purifying agent, a feed or a feed additive. A fifth object of the present invention is to provide a product for controlling infection of Vibrio candidum or Vibrio anguillarum, which contains the phage Vibrio campbellii phage vB _ VcaS _R45Z or microbial preparation as described above. The sixth object of the present invention is to provide a method for controlling Vibrio paravaliensis or Vibrio anguillarum harmful infection in vitro, which kills Vibrio paravaliensis or Vibrio anguillarum in vitro by using the above-mentioned cleavage effect of phage Vibrio campbellii phage vB _ VcaS _r45z or microbial preparation on Vibrio paravaliensis or Vibrio anguillarum. The system provided by the invention analyzes the biological characteristics, cleavage kinetics and genome structure and system classification status of phage R45Z pair Vibrio campbellii JL 2918. The results of the lysis kinetics show that R45Z can trigger lysis at the early proliferation stage of bacteria at different infection doses, and the antibacterial effect of the bacteria is obvious in MOI dependence and time dependence. The high MOI group has 3-4 h after infection even though the bacterial density is obviously reduced, and the medium-low MOI group has weaker antibacterial degree, but can keep effective inhibition on the host in the later period of action. The one-step growth curve shows that R45Z has a latency of about 20 min and a high burst, indicating that it is characterized by rapid replication and high cleavage efficiency. The whole genome sequencing result shows
