In addition, the regulation of hsa_circ_0005737 expression can influence the osteogenic differentiation capacity and inflammatory response of periodontal ligament stem cells, indicating that the hsa_circ_0005737 can be used as a potential target for periodontitis treatment. The invention also provides a primer sequence, a detection reagent combination and a kit for detecting hsa_circ_0005737, and provides a new solution for noninvasive, early and high-specificity diagnosis and targeted treatment of periodontitis.

- 1. A periodontitis marker is characterized in that the marker is a circular RNA hsa_circ_0005737, and the nucleotide sequence of the circular RNA hsa_circ_0005737 is shown as SEQ ID NO. 1.
- 2. Use of the circular RNA hsa_circ_0005737 of claim 1 for the preparation of a product for diagnosing periodontitis.
- 3. Use of the circular RNA hsa_circ_0005737 of claim 1 for the preparation of a product for assessing periodontitis activity, severity or prognosis.
- 4. Use of the circular RNA hsa_circ_0005737 of claim 1 for the preparation of a product for monitoring the efficacy of treatment of periodontitis.
- 5. A reagent combination for periodontitis diagnosis, prognosis evaluation and/or efficacy monitoring, which is characterized in that the reagent combination comprises a reagent for specifically detecting the expression level of hsa_circ_0005737 in a sample, and the nucleotide sequence of the circular RNA hsa_circ_0005737 is shown as SEQ ID NO. 1.
- 6. The reagent combination according to claim 5, wherein the reagent comprises a primer pair for specifically amplifying circular RNA hsa_circ_0005737, the primer pair sequences of which are shown in SEQ ID NO. 4 and SEQ ID NO. 5.
- 7. The combination of reagents according to claim 5 or 6, wherein the sample is one or more clinical biological samples selected from the group consisting of serum, plasma, whole blood, saliva, gingival crevicular fluid, buccal swab, pus, body fluid, periodontal tissue, paraffin section.
- 8. A kit for the diagnosis, prognosis and/or monitoring of the efficacy of a treatment of periodontitis, characterized in that it comprises a combination of reagents according to any one of claims 5 to 7.
- 9. A pharmaceutical composition for preventing, alleviating and/or treating periodontitis, characterized in that it comprises an active ingredient capable of down-regulating the expression level of the circular RNA hsa_circ_0005737 of claim 1.
Description
Periodontitis marker and application thereof Technical Field The invention relates to the technical field of biomedical detection technology and plant quarantine, in particular to a periodontitis marker and application thereof. Background Periodontitis is a chronic inflammatory disease initiated by plaque biofilm and is characterized by progressive destruction of periodontal support tissues (including gums, periodontal membranes, alveolar bone, and cementum), which ultimately leads to loosening and loss of teeth. Periodontitis is a major cause of adult tooth loss worldwide and is closely associated with a variety of systemic diseases (e.g., diabetes, cardiovascular disease, rheumatoid arthritis, etc.). Currently, clinical diagnosis and assessment of periodontitis is mainly dependent on traditional clinical examination methods such as depth of investigation, clinical adhesion loss, bleeding index and imaging examinations (e.g., X-ray films). However, these methods have significant limitations: The subjectivity is strong, the clinical examination result is influenced by the experience of an operator, and the tissue damage is mainly reflected, so that early warning of diseases and accurate judgment of the active period cannot be realized. The method has the advantages of invasiveness and limitation that the imaging examination has radiation exposure and is difficult to frequently carry out, the microorganism detection can identify pathogenic bacteria, but the method has complex operation, long time consumption and high cost, the oral flora is complex, and the relevance between the detection of single or multiple pathogenic bacteria and the disease activity is not absolute. The body fluid markers have insufficient specificity, namely certain inflammatory factors (such as IL-1 beta, IL-6, TNF-alpha, MMPs and the like) in gingival crevicular fluid or saliva are researched as potential markers, but the level of the body fluid markers is easily influenced by other oral or systemic inflammations, the specificity is not high, and the stability is poor. Therefore, the development of a novel biomarker which can realize early, noninvasive, high-specificity and high-sensitivity diagnosis and effectively distinguish the active period from the stationary period of diseases and evaluate the curative effect and prognosis is a key problem to be solved in the field of periodontitis clinical diagnosis and treatment. Circular RNAs (circrnas) are a class of non-coding RNA molecules that do not have a 5 'cap and a 3' poly (a) tail, forming a closed circular structure by covalent bonds. Because its loop structure is not easily degraded by exonucleases, circRNA is more stable in vivo than linear RNA. Recent studies have shown that circRNA has a tissue/disease-specific expression pattern and can be involved in the regulation of cellular physiopathological processes by acting as microRNA (miRNA) sponge, interacting with RNA binding proteins, regulating gene transcription or translation, etc. These properties make circRNA a very potential new generation of disease diagnostic biomarkers and therapeutic targets. However, no specific circRNA is currently used as a specific diagnostic and prognostic evaluation marker for periodontitis and is used for guiding a clear report of treatment, so that development of a circRNA marker capable of realizing early, noninvasive, high-specificity and high-sensitivity diagnosis and effectively distinguishing active period from stationary period and evaluating curative effect and prognosis of diseases is a technical problem to be solved urgently by those skilled in the art. Disclosure of Invention In view of this, the present invention has developed a periodontitis marker and its application. The first aim of the application is to provide a periodontitis marker, wherein the marker is circular RNA hsa_circ_0005737, and the nucleotide sequence of the marker is shown as SEQ ID NO. 1. It is a further object of the present application to provide the use of the circular RNA hsa_circ_0005737 in the preparation of a product for diagnosing periodontitis. It is a further object of the present application to provide the use of the circular RNA hsa_circ_0005737 in the preparation of a product for assessing periodontitis activity, severity or prognosis.
