Claims (13)
- A compound of Formula (I) N-oxide, or a salt thereof, wherein: X is and Y is S; or X is S and Y is N or CH; R 1 is independently -CH 3 , -CH 2 CH 3 , or -CH(CH 3 ) 2 ; G is: each R 2 is independently -CN, -CH 3 , or -OCH 3 ; A is: (i) -CH 2 NHCH 2 C(O)NH 2 ; (ii) -CH 2 R 3 , -CH 2 NHR 3 , -CH 2 NHCH 2 R 3 , -C(O)NHR 3 , -C(O)NR x R 4 , or -C(O)R 3 ; or (iii) cyclohexyl, piperidinyl, piperazinyl, methylpiperazinyl, dimethylpiperazinyl, or diazaspiro[3.3]heptanyl, each substituted with zero to 1 R 3a ; R 3 is azetidinyl, oxetanyl, cyclopropyl, cyclobutyl, pyrrolidinyl, pyrrolidinonyl, piperidinyl, piperazinyl, piperazinonyl, morpholinyl, dioxidothiomorpholinyl, oxazepanyl, diazaspiro[3.3]heptanyl, dioxidothiaazaspiro[3.3]heptanyl, oxaazaspiro[3.3]heptanyl, or oxaazaspiro[3.5]nonanyl, each substituted with zero to 1 R 3a and zero to 2 R 3b ; R 3a is F, -CH 3 , -CH 2 CH 3 , -CH 2 CH 2 CH 3 , -CH(CH 3 ) 2 , -CH 2 CH(CH 3 ) 2 , -CH 2 C(CH 3 ) 3 , -CH 2 CH(CH 2 CH 3 ) 2 , -CH 2 CF 3 , -CH 2 CN, -CH 2 C(CH 3 ) 2 OH, -CH 2 CH 2 OCH 3 , -CH 2 CH 2 S(O) 2 CH 3 , -CH 2 C(O)NR x R x , -OCH 3 , -C(O)CH 3 , -C(O)CH 2 N(CH 3 ) 2 , -NH 2 , -NH(CH 3 ), -NH(CH(CH 3 ) 2 ), -NH(CH 2 C(CH 3 ) 3 ), -NH(CH 2 C(CH 3 ) 2 OH), -NH(CH 2 CHFC(CH 3 ) 2 OH), -NH(CH 2 C(CH 3 ) 2 OCH 3 ), -N(CH 3 )(CH 2 CH 3 ), -N(CH 3 )(CH(CH 3 ) 2 ), -N(CH 3 )CH 2 CH(CH 3 ) 2 , -N(CH 3 )CH 2 C(O)N(CH 3 ) 2 , -N(CH 3 )CH 2 CH 2 S(O) 2 CH 3 , -NHC(O)CH 2 N(CH 3 ) 2 , -NHC(O)CH 2 N(CH 3 )(CH 2 CH 3 ), -NHC(O)CH 2 NH(CH 2 CH(CH 3 ) 2 ), -NHCH 2 C(O)NH 2 , -NHCH 2 C(O)NH(CH 3 ), -NHCH 2 C(O)N(CH 3 ) 2 , -NHCH 2 CH 2 NH(CH 3 ), -NHS(O) 2 CH 3 , -S(O) 2 CH 3 , -CH 2 (cyclopropyl), -CH 2 (methyloxetanyl), -CH 2 (tetrahydropyranyl), -C(O)(dioxidotetrahydrothiopyranyl), -NH(cyanocyclopropyl), -NH(cyclobutyl), -NH(methylcyclobutyl), -NH(oxetanyl), -NH(methyloxetanyl), -NH(ethyloxetanyl), -NH(isopropylpiperidinyl), -NH(tetrahydropyranyl), -NH(bicyclo[1.1.1]pentyl), -NHCH 2 (methyloxetanyl), -NH(dioxidotetrahydrothiophenyl), -NHCH 2 (hydroxymethylcyclopropyl), -NHCH 2 (methylsulfonylcyclopropyl), -NHCH 2 (methylcyclobutyl), -NHCH 2 C(O)(morpholinyl), cyclobutyl, azetidinyl, fluoroazetidinyl, difluoroazetidinyl, hydroxyazetidinyl, methoxyazetidinyl, hydroxy(trifluoromethyl)azetidinyl, bis(hydroxymethyl)azetidinyl, oxetanyl, pyrrolidinyl, acetamidopyrrolidinyl, piperazinonyl, tetrahydropyranyl, morpholinyl, dioxothiomorpholinyl, oxazepanyl, bicyclo[1.1.1]pentanyl, azaspiro[3.3]heptanyl, oxaazaspiro[3.3]heptanyl, dioxidothiaazaspiro[3.3]heptanyl, oxaazabicyclo[3.2.1]octanyl, oxaazaspiro[3.5]nonanyl, or diazaspiro[4.4]nonanyl; each R 3b is -CH 3 ; R 4 is H, -CH 2 CHF 2 , -CH 2 CF 3 , -CH 2 CH 2 N(CH 3 ) 2 , -CH 2 R 3 , -CH 2 CR x R x R 3 , or -(CH 2 ) 3 R 3 ; R 5 is H or -CH 2 CF 3 ; each R x is independently H or -CH 3 ; and p is zero, 1, or 2.
- The compound according to claim 1, N-oxide, or a salt thereof, wherein X is N and Y is S.
- The compound according to claim 1, N-oxide, or a salt thereof, wherein A is cyclohexyl, piperidinyl, piperazinyl, methylpiperazinyl, dimethylpiperazinyl, or diazaspiro[3.3]heptanyl, each substituted with zero to 1 R 3a .
- The compound according to claim 1, N-oxide, or a salt thereof, wherein A is -CH 2 R 3 , -CH 2 NHR 3 , -CH 2 NHCH 2 R 3 , or -CH 2 NHCH 2 C(O)NH 2 .
- The compound according to claim 1, N-oxide, or a salt thereof, wherein A is -C(O)NHR 3 or -C(O)NR x R 4 .
- The compound according to claim 1, N-oxide, or a salt thereof, wherein A is -C(O)R 3 .
- The compound according to claim 1, N-oxide, or a salt thereof, wherein X is S and Y is N.
- The compound according to claim 1, N-oxide, or a salt thereof, wherein X is S and Y is CH.

CROSS REFERENCE This application claims priority from U.S. Provisional Application Serial No. 62/908,840 filed October 1, 2019. DESCRIPTION The present invention generally relates to substituted bicyclic heteroaryl compounds useful as inhibitors of signaling through Toll-like receptor 7, 8, or 9 (TLR7, TLR8, TLR9) or combinations thereof. Provided herein are substituted bicyclic heteroaryl compounds, compositions comprising such compounds, and methods of their use. The invention further pertains to pharmaceutical compositions containing at least one compound according to the invention that are useful for the treatment of conditions related to TLR modulation, such as inflammatory and autoimmune diseases, and methods of inhibiting the activity of TLRs in a mammal. Toll/IL-1 receptor family members are important regulators of inflammation and host resistance. The Toll-like receptor family recognizes molecular patterns derived from infectious organisms including bacteria, fungi, parasites, and viruses (reviewed in Kawai, T. et al., Nature Immunol., 11:373-384 (2010)). Ligand binding to the receptor induces dimerization and recruitment of adaptor molecules to a conserved cytoplasmic motif in the receptor termed the Toll/IL-1 receptor (TIR) domain with the exception of TLR3, all TLRs recruit the adaptor molecule MyD88. The IL-1 receptor family also contains a cytoplasmic TIR motif and recruits MyD88 upon ligand binding (reviewed in Sims, J.E. et al., Nature Rev. Immunol., 10:89-102 (2010)). Toll-like receptors (TLRs) are a family of evolutionarily conserved, transmembrane innate immune receptors that participate in the first-line defense. As pattern recognition receptors, the TLRs protect against foreign molecules, activated by pathogen associated molecular patterns (PAMPs), or from damaged tissue, activated by danger associated molecular patterns (DAMPs). A total of 13 TLR family members have been identified, 10 in human, that span either the cell surface or the endosomal compartment. TLR7/8/9 are among the set that are endosomally located and respond to single-stranded RNA (TLR7and TLR8) or unmethylated single-stranded DNA containing cytosine-phosphate-guanine (CpG) motifs (TLR9). Activation of TLR7/8/9 can initiate a variety of inflammatory responses (cytokine production, B cell activation and IgG production, Type I interferon response). In the case of autoimmune disorders, the aberrant sustained activation of TLR7/8/9 leads to worsening of disease states. Whereas overexpression of TLR7 in mice has been shown to exacerbate autoimmune disease, knockout of TLR7 in mice was found to be protective against disease in lupus-prone MRL/lpr mice. Dual knockout of TLR7 and 9 showed further enhanced protection. As numerous conditions may benefit by treatment involving modulation of cytokines, IFN production and B cell activity, it is immediately apparent that new compounds capable of modulating TLR7 and/or TLR8 and/or TLR9 and methods of using these compounds could provide substantial therapeutic benefits to a wide variety of patients. WO 2019/126253 discloses aryl and heteroaryl substituted indole compounds that act as inhibitors of signaling through Toll-like receptor 7, or 8, or 9. WO 2019/125977 discloses 4-azaindole compounds that act as inhibitors of signaling through Toll-like receptor 7, or 8, or 9. Swarnali et al., European Journal of Medicinal Chemistry, 134 (2017), 334-347, discloses benzoxazole derivatives with Toll-like receptor 9 antagonism. SUMMARY OF THE INVENTION The present invention relates to a new class of substituted bicyclic heteroaryl compounds found to be effective inhibitors of signaling through TLR7/8/9. These compounds are provided to be useful as pharmaceuticals with desirable stability, bioavailability, therapeutic index, and toxicity values that are important to their drugability. The present invention provides compounds of Formula (I) that are useful as inhibitors of signaling through Toll-like receptor 7, 8, or 9 and are useful for the treatment of proliferative diseases, allergic diseases, autoimmune diseases and inflammatory diseases, or stereoisomers, N-oxides, tautomers, pharmaceutically acceptable salts, or solvates thereof. The present invention also provides pharmaceutical compositions comprising a pharmaceutically acceptable carrier and at least one of the compounds of the present invention or stereoisomers, tautomers, pharmaceutically acceptable salts, or solvates thereof. The compounds of the present invention may be used in a method for inhibition of Toll-like receptor 7, 8, or 9 comprising administering to a host in need of such treatment a therapeutically effective amount of at least one of the compounds of the present invention or stereoisomers, tautomers, pharmaceutically acceptable salts, or solvates thereof.
