Abstract

- In paragraph 1, A pharmaceutical composition for the prevention, treatment, or improvement of symptoms of male menopause, characterized in that the above strain includes its live cells, dead cells, cultures, culture medium containing cells, culture medium from which cells have been removed, concentrates of culture medium, and metabolites isolated from cells or culture medium.

Description
Composition containing Streptococcus thermophilus ATG-V11 strain for improving andropause The present invention relates to a composition for improving symptoms of male menopause containing the strain Streptococcus thermophilus ATG-V11 ( Streptococcus thermophilus ATG-V11, KCTC15543BP). The term menopause has traditionally been used exclusively for women, but recently, this concept has emerged in men as well, as endocrine changes resulting from the gradual decrease in male hormone production occur with aging. In men in their 40s and 50s, male hormone secretion gradually decreases, and in their 70s, male hormone secretion drops to half the level of their 30s, while the sensitivity of target cells to testosterone decreases. Consequently, the appearance of various menopausal symptoms similar to those seen in women is referred to as male menopausal symptoms (Partial Androgen Deficiency of Aging Male, PADAM; androgen deficiency for aging male, ADAM; Male climateric, Andropause). The main symptoms of male menopause include easy fatigue, memory loss, and frequent symptoms of depression. Physically, it manifests as decreased muscle strength, increased body fat, and weakened bones. Sexual function also declines, leading to issues such as erectile dysfunction and decreased libido. Major causes include a decrease in testosterone due to the aging of the brain and testes, environmental factors such as excessive alcohol consumption, smoking, and stress, and physical factors such as hypertension, diabetes, and liver disease. Furthermore, as the activity of TM3 cells (mouse reproductive cells, testosterone-producing cells), a type of male germ cell, decreases, the rate of TM3 cell death also increases. Additionally, the expression of PDE (phosphodiesterase) protein, which is associated with erectile dysfunction, increases. Meanwhile, to treat male menopausal syndrome, improvement effects have been confirmed using male hormone replacement therapy such as testosterone undecanoate, but as side effects from hormone therapy can affect the liver, lipid status, cardiovascular and prostate diseases, recently, attempts are being made to develop male menopausal syndrome improvement agents with natural products or fermented products as main ingredients. Accordingly, the inventors confirmed that the novel Streptococcus thermophilus ATG-V11 strain increases the expression of testosterone, and confirmed that it can be used as a composition derived from natural products for improving male menopause, thereby completing the present invention. Preferred embodiments of the present invention will be described in detail below. However, the present invention is not limited to the embodiments described herein and may be embodied in other forms. Rather, the content introduced herein is provided to fully convey the concept of the present invention to those skilled in the art, so that it may be thorough and complete. <Example 1. Isolation and Identification of Novel Lactic Acid Bacteria - ATG-V11> 90 ml of physiological saline (0.85% NaCl/L) was placed in an autoclaved bag, 10 g of homemade yogurt collected from the Daejeon area was added and mixed uniformly, and 1 ml was dispensed and serially diluted 10-fold in 9 ml of physiological saline and plated three times on BL solid medium. Afterward, the cultures were incubated in a 37°C incubator for more than 48 hours, and the grown bacteria were analyzed using a catalase test with 0.3% hydrogen peroxide and Gram staining. Subsequently, catalase-negative, Gram-positive, rod-shaped bacteria were selected, and among them, strains with high testosterone secretion ability were isolated. The 16S rRNA gene sequences of the two isolated strains were analyzed by Macrogen, and the sequences obtained through 16S rRNA sequencing were compared with the NCBI BLAST database. As a result, the 16S rRNA sequences of these strains matched those of Streptococcus thermophilus by 99.9%, indicating that they belong to the taxonomic genus Streptococcus thermophilus and were identified as novel strains having the gene sequences show
