Abstract

The present invention relates to a method for preparing a coffee silver skin extract that enhances functional components and efficacy by utilizing coffee silver skin, which is known to contain many beneficial substances with functionalities such as improving muscle and cardiovascular diseases, as well as anti-diabetic, antioxidant, anti-inflammatory, and anti-obesity properties. More specifically, the extract obtained by micro-grinding coffee silver skin and then extracting it with a specific enzyme has the effect of enhancing the content and functionality of polyphenols and chlorogenic acid, and can be usefully utilized as a material for health functional foods.

Claims (7)

  1. (1) A step of micro-grinding coffee silver skin; (2) A step of preparing a coffee silver skin solution by adding water to the micro-ground coffee silver skin of step (1) above; and (3) A method for preparing a coffee silver skin extract characterized by including the step of adding cellulase to the coffee silver skin solution prepared in step (2) above and then enzymatically treating it.
  2. A method for producing coffee silver skin extract according to claim 1, wherein the micro-grinding in step (1) is micro-grinding to an average particle size of 50 to 100 μm.
  3. In paragraph 1, (1) A step of micro-grinding coffee silver skin to an average particle size of 50 to 100 μm; (2) A step of preparing a coffee silver skin solution by adding 40 to 60 times (v/w) of water relative to the coffee silver skin to the micro-ground coffee silver skin of step (1) above; and (3) A method for preparing a coffee silver skin extract characterized by including the step of adding cellulase to the coffee silver skin solution prepared in step (2) above and then enzymatically treating it at 35 to 45°C for 20 to 30 hours.
  4. A method for preparing a coffee silver skin extract according to any one of claims 1 to 3, wherein the method for preparing the coffee silver skin extract is characterized by enhancing the polyphenol and chlorogenic acid content and antioxidant activity of the coffee silver skin extract.
  5. Coffee silver skin extract prepared by any one of the methods of paragraphs 1 to 4.
  6. An antioxidant health functional food composition comprising the coffee silver skin extract of claim 5 as an active ingredient.
  7. A health functional food composition for the prevention or improvement of diabetes, muscle disease, or cardiovascular disease comprising the coffee silver skin extract of claim 5 as an active ingredient.

Description

Method for producing functional extract using coffee silver skin and health functional food composition using the same The present invention relates to a method for preparing a coffee silver skin extract and a coffee silver skin extract prepared by said method. The coffee silver skin extract of the present invention has enhanced active ingredient content and functional effects, and thus relates to a technology that can be usefully applied in the fields of health functional foods and processed foods. Coffee silver skin is a silvery outer layer that surrounds the center cut and the endosperm (green bean) of a coffee bean, and is also called silver skin. Outside the coffee silver skin is an endocarp called parchment, and beyond that is a flesh called pulp. The outermost layer is an exocarp called the outer skin. The silverskin naturally peels off and is removed during the coffee roasting process. Since the quality of the coffee's flavor deteriorates if the silverskin is not properly removed from the beans, the silverskin generated during the coffee processing is generally disposed of as waste, or some is used as fertilizer. Coffee silver skin is known to be rich in antioxidants, polysaccharides, and polyphenols, which enhance immunity and provide anti-inflammatory effects; as a result, it is garnering attention due to the recent rise in interest in health and well-being. Furthermore, coffee silver skin has been reported to possess antibacterial properties against harmful bacteria such as K. pneumoniae , S. epidermidis , and S. aureus . In addition, coffee silver skin is effective in strengthening muscles and preventing sarcopenia, and can contribute to maintaining the health of the elderly in particular. Therefore, coffee silver skin is attracting attention as a source of industrially usable functional substances. In particular, as demand for functional foods for the elderly increases in various countries, including Japan, it is expected to provide new market opportunities, and thus the utilization of silver skin is likely to receive even more attention. Accordingly, the present invention has developed a method to obtain a high-quality coffee silver skin extract that can be utilized as a diverse food material, with enhanced extraction efficiency and nutritional content, and excellent palatability, by utilizing coffee silver skin, which is mainly discarded. Regarding prior art utilizing conventional coffee silver skin, Korean Published Patent No. 2019-0088424 discloses a composition in which an ethanol-extracted extract of coffee silver skin is used in the production of alcoholic beverages, and Korean Published Patent No. 2021-0090543 discloses a composition for the prevention or treatment of muscle diseases comprising an ethanol-extracted extract of coffee silver skin; however, there is no technology for producing an extract by micro-grinding and enzyme complex treatment of coffee silver skin as in the present invention. Figure 1 is a graph comparing the polyphenol content of coffee silver skin extract. Figure 2 is a graph comparing the chlorogenic acid content of coffee silver skin extract. Figure 3 is a graph comparing the DPPH radical scavenging activity of coffee silver skin extracts. Comparative Example 1 of FIGS. 1 to 3 refers to: ethanol extract, Comparative Example 2 refers to: hot water extract, Comparative Example 3 refers to: enzyme extract, Comparative Example 4 refers to: micro-grinding + ethanol extract, Comparative Example 5 refers to: micro-grinding + hot water extract, and Preparation Example 1 refers to: micro-grinding + enzyme extract. In addition, other superscripts in Figures 1 to 3 indicate a statistically significant difference, p<0.05.
