Description

Composition for anti-wrinkle and enhancing skin elasticity and its use The present invention relates to a composition for improving skin wrinkles and enhancing skin elasticity, and more specifically, to a composition for improving skin wrinkles and enhancing skin elasticity containing as an active ingredient a complex extract obtained by mixing bamboo stem, thistle whole plant and Chinese yam in a specific mixing ratio, in particular a cosmetic composition and a health functional food. Human skin protects the body from external physical and chemical stimuli and is broadly divided into three layers: the dermis, epidermis, and subcutaneous fat tissue. The dermis is primarily composed of fibroblasts, which mainly synthesize collagen and other proteins while producing small amounts of lipids. Among these, matrix proteins exist in the dermal layer and serve as the components filling the spaces between cells; they consist largely of collagen, which accounts for over 90%, and elastin, which accounts for 3–4%. Collagen, which makes up the majority of the skin's dermis, and elastin, which possesses high elasticity, are major proteins produced by fibroblasts and are known to play a significant role in skin elasticity. Matrix proteins are primarily produced by fibroblasts and decrease by 1% annually with aging. The weakening of skin cells due to a decrease in matrix proteins in the skin leads to a decline in skin elasticity. One of the factors contributing to the decline in the dermis layer, which is the most important element in skin elasticity loss, is the reduced production of proteins such as collagen and elastin, which are produced by fibroblasts and form the majority of connective tissue. Matrix proteins are degraded by collagenase, elastase, and the like. The expression of these collagen and elastin-degrading enzymes is the primary cause of wrinkle formation in the skin (Published Patent No. 10-2017-0037570). Currently, there are four types of functional ingredients for wrinkle improvement notified by the Ministry of Food and Drug Safety: retinol, retinyl palmitate, adenosine, and polyethoxylated retinamide. However, there are significant issues regarding side effects, such as irritation and inflammation, as much as there are effects. Driven by the growing awareness of beauty and health and the emphasis on environmental protection, there is a strong trend toward reducing the use of chemical ingredients in cosmetics. Consequently, interest in products utilizing natural extracts is on the rise. Natural extracts are ingredients derived from nature, such as plants; they possess the advantage of being chemically safe, capable of providing functional benefits to the skin, and having a low probability of causing side effects. Therefore, there is a continuing demand for the development of compositions for improving skin wrinkles and enhancing skin elasticity using natural extracts with fewer side effects on the skin, instead of synthetic raw materials. The present invention is further explained in detail by the following examples. These examples are intended to illustrate the invention and the scope of the invention should not be limited by them. Preparation Example 1: Preparation of a complex extract Bamboo stems, thistle whole plant, and yam were purchased from Dongkwang Natural Farm Co., Ltd. in a dried state and powdered to prepare each powder. Each powder was mixed in the weight ratios shown in Table 1 below. weight ratiobamboo stemThistle whole plantWild yamExample 1121Comparative Example 1211Comparative Example 2112Comparative Example 3111 1 L of 50% fermented alcohol was added to 100 g of each mixed powder and extracted using an ultrasonic extractor at room temperature (20~30℃) for 2 hours, and then filtered through filter paper to prepare a complex extract. The complex extract was freeze-dried for storage and used in the following analysis tests. Test Example 1: Analysis of Elastase Inhibitory Activity Elastase is known to be a major cause of skin elasticity loss and wrinkle formation by breaking down elastin, a protein that maintains skin elasticity within the dermis. It is known that inhibiting the activity of elastase can improve skin wrinkles. The elastase inhibitory activity analysis was performed by using N-succinyl-L(ala)3-p-nitroanilide as a substrate and measuring the amount of p-nitroanilide produced from the substrate. Specificall
